Cerebrovascular diseases do great harm to human's heath, which isone of the important reasons leading to disability and death. Reperfusioninjury after cerebral ischemia has become the key problem in the clinicaltherapy of ischemic cerebrovascular disease. It is reported that thehawthorn leaves flavonoids have obvious efficiency on reducing, bloodpress, protectcting cradrac muscle with meager blood and controllingblood platelet agglomerating. Furthermore, they have potent effects onantiaging, anticancer andcan enhance organism immunity. The significanceof the experiment lies in that through establishing the models of rats'reperfusion injury after cerebral ischemia, observing the protectiveexpression after the intervention of hawthorn leaves flavonoids'application.The experiment use the string-fastening method to establish focalischemia-reperfusion models in Wistar rats, we measure the LDH and CK inserum, SOD and MDA are measured in rat brains. TTC-dying method is appliedto determine the volumes of cerebral infarctions and nerve function gradesscoring to estimate the behaviour of the model. The result demonstratedthat compared with model group, brain contents of MDA decreasedsignificantly and those of SOD increased significantly in HLF group. AndHLF group reduce LDH and CK activities in serum. The experiment confirmesthat the HLF can increase the activity of SOD in the brain tissue, andcan prohibit the higher MDA, correspondingly decline LDH and CK activityin serum. Because the SOD is the best thing to clear free radical formation,MDA is the last thing in the progress of free radical formation reducedthe fat sufoxygen. So HLF have the function of releasing free radicalformation, contending with fat over oxidizing and protecting brain nerve.The experiment also confirmes that HLF diminish cerebral infarction areaand neurological deficit scores in the ischemic-reperfusion rats indosage dependently, thus they play the roles of cerebral protection.Matrix metalloproteinases are one of the most important proteolytic enzymes that degrade the extracellular matrix. After brain ischemia andreperfusion, there is an increase of MMPs, in particular the activationincrease of MMP-9 is associated with vascular permeability, blood-brainbarrier permeability, inflammatory cell influx and brain edema. MMP-9immunohistochemistry showed in the sham operated group no MMP-9expressions could been seen in neuron necrosis, in the model groupMMP-9's expreesion immunopositive, while hawthorn leaves flavonoidstreatment group compared with the sham operated group and model group,MMP-9's immunopositive expressions were situated between the two groups.So we conclude that the hawthorn leaves flavonoids can release the brainedema which the ischemia-induced brain damage reduce, and finallysuppressing their nerve toxicity.Protective mechanisms of hawthorn leaves flavonoids forischemia-reperfusion cerebral injury are futher discussed and proposetheoretical basis for use of them though our experiment, from these partsthat anti-oxidase injure, reduce brain edema and decrease brainmicrocirculation. In the subsequent experiment and clinical work, We willdeeply discuss the function and it's new therapy strategy to providesufficient experimental evidence for its clinical application in treatingischemic cerebrovascular disease.
