Experimental Study On The EAU Induced With The Retinal And Non-retinal Antigen

Objective: The purpose of this study is to investigate the experimentalautoimmune uveoretinitis (EAU) induced with the human soluble-antigen peptide 35(HS-AgP35) and experimental autoimmune anterior uveitis(EAAU) induced with guinea pig myelin basic protein peptide(gp-MBP20),and study the immune mechanism of the different type of autoimmunity uveitis.Methods: All Lewis rats, except controls, were immunized with BS-Ag,HS-AgP35 or gp-MBP20 emulsified with equal volume complete Freund's adjuvant (CFA);Clinical disease was monitored daily by grading signs of EAU over 14 days after the second EAU induction. At day 21, all Lewis rats were killed and their eyes were removed for histopathological examination, serum was collected and tested in ELISA for the presence of antibodies against retinal BS-Ag and HS-AgP35, spleens were also removed for RT-PCR analysis of IL-4 mRNA 和 IFN-γ mRNA expression.Results: (1) All antigens can induced the uveitis in Lewis rats, but the incidence ofthe rats immunized with HS-AgP35 or gp-MBP20 is 100% and no difference(P>0.05), and the time of duration of the rats immunized with HS-AgP35 is longer and its severity of disease is much worse than the rats immunized with BSA-g (P<0.05) . (2) there was significantly difference in the level of serum antibodies among all groups (P<0.05 ) . (3) As being expected, there wassignificantly difference in the level of spleen IL-4 mRNA $i IFN-y mRNA expression among the nomah the rats immunized with HS-AgP35 or BS-Ag (P<0.05), but the rats immunized with HS-AgP35 is no different from the rats immunized withgp-MBP20(P>0.05).Conclusions: 1. The EAU acquired from the Lewis rats induced with HS-AgP35is much better than it acquired from BS-Ag. So this study affords a method of establishing a higher incidence and severer EAU to study the immunity uveitis. 2. The gp-MBP20 is also good autoantigen for establishing a higher incidence and severer EAAU. Different autoantigen induces the inflammations in the eyes are different.This study will help us to find the exactly autoantigen induced the autoimmune uveitis, and offered a immunologial basic for the treatment of the uveitis by oral tolerance.Key words: S-Ag;myelin basic protein/MBP;experimental autoimmunity uveitis/EAU...
