ObjectiveTo evalate the liability of Bone Mesenchymal stem cells(BM-SCs),the effects of the cardiac muscle cells multiplication,blood vessel regeneration and theheart function of the Chinese traditional medicine XinFuKang(XFK)combiningBMSCs transplantation ,and to discuss the mechamism of XFK on Acute myocardial infarction(AMI) rats.MethodsBMSCs cells were collected from bone marrow of Wistar rat's femoral bo nes,cultured and passaged in vitro,then purified by gradient centrifugation - ad hesive method. The animal with acute myocardial infarction were built by cor onary artery ligation.The Wistar rats were randomly divided into 7groups:(1)sh am peration groups(S group,n=9);(2).control group(C group n=11),ligate the cor onary artery;(3)low dose of XFK group(L group,n=12):therapy with XFK 40.5 g/kg/d in two weeks;(4)middle dose of XFK group(M group,n=10): therapy wi th XFK 81g/kg/d in two weeks;(5)BMSCs transplantation group(B group,n=8):t rantplant BMSCs 2×10~7/ml signed with BrdU;(6)BMSCs transplantion combin ed with low dose of XFK(LB group,n=8):trantplant BMSCs 2×10~7/ml signed with BrdU and treated with XFK 40.5g/d/kg;(7)BMSCs transplantion combined with middle dose of XFK(LB group,n=8):trantplant BMSCs 2×10~7/ml signed with BrdU and treated with XFK 81g/d/kg.At the end of two weeks,exami ned the rats' heart function,evaluted the infacrted area with N-BT, and the lialiability of BMSCs, the multiplication of cardiac musclecells,the regeneration of blood ve-ssles by immunohistochemistry of BrdU, PCNA and CD34.Results:1.Compared with the C group,each therapy group's infarct area declined;the yentricle thickness of infarct increased;the heart func-tion improved.2.1n the margian of infarct of B group,LB group and MB group ,there wereBrdU* cells, and the amunt of BrdU* cells of LB, MB group were more compared with B grooup.3.Compared with the C group,the amount of CD34+ capillary vessels of eachtherapy group were increased;and compared with B group, the amount of MB group's capillary vessels were increased.4. Compared with the C group, the amount of PCNA+ cells of each therapygroup were increased;and compared with B group.the amount of PCNA+ cellswere increased in traditional medicine therapy groups and groups treated withcombined therapy.Conclusion:On AMI rats,each therapy method can reduced the infarct area,increaed th e thickness of infarct ventricle ,accelerated blood vessles' regeneration,improve d the heart function. Traditional medicine can promote the multiplication of ca rdiac muscle cells,and the effects were preceded with pure BMSCs transplantat ion. The effects concerning the increase of BMSCs livability,the promotion of blood vessles regeneration, the improvement of heart funtion and the cardiacmuscle multiplication, the combination therapy method were better than pure BMSCs transplantation.
