Objective:To study the therapeutic effect of compound traditional Chinese medicine on rat model of liver fibrosis induced by fat-rich diet.Methods:Fourty-two male SD rats were randomly divided into 4 groups accoding to the weight: control group(n=8),fed normal diet;model group(n=18);traditional Chinese medicine high dose group(n=8);traditional Chinese medicine low dose group(n=8), fed fat-rich diet.Four of them were randomly killed after 24 weeks to be identified the model of liver fibrosis.Low dose group and high dose group were perfused with Chinese herbs for 12 weeks.Model group and control group were perfused with NS.All rats were weighted every week,after 12 weeks of treatment,all rats were killed.We examined the effect of traditional Chinese medicine on body weight,liver weight,ALT,AST,TC,TG, TGF β1 in serum and hepatic tissue.The hepatic histologic change was evaluated by light microscopy. Results:l.Body weight and liver weight: The levels of body weight were significantly higher in model group and two treatment groups than those in control group,were significantly lower in two treatment groups than those in model group.The levels of liver weight in model group, high dose group and low dose group were higher than those in control group. The levels of two treatment groups were lower than those of model group. 2.Serum biochemical parameters:The levels of TC,TG,ALT,AST in serum weresignificantly higher in model group and two treatment groups than those in controlgroup;The levels of TC,ALT,AST in serum were significantly lower in two treatmentgroups than those in model group;The levels of TG were higher in model group thanthose in high dose group.3.HA, LN, PCⅢ, C IV:The levels of HA, LN, PCⅢ, C IV in serum were higherin model group than those in the rest three groups, were higher in low dose groupthan those in control group,There was no difference between high dose group andcontrol group.4.TGF β 1 :The expressions of TGF β 1 in hepatic tissue were lower in control groupthan those in model group;were lower in high dose group than those in modelgroup;There was no difference between low dose group and model group.5.Histology:The levels of steatosis,steatohepatitis and fibrosis were significantlylower in control group than those in model and low dose group;were significantlylower in high dose group than those in model group.There was no difference betweenmodel group and low dose group.Conclusion:1.The therapeutic effect of compound tranditional Chinese medicine is remarkable onrat model of liver fibrosis induced by fat-rich diet.2.Compound tranditional Chinese medicine is safe to liver.3. Compound tranditional Chinese medicine can low the levels of TC,TG,ALT,AST,HA,LN,PCIII,CIV in serum,protect liver cells,improve liver function,inhibit theproduction of collagen and improve it's degration.inhibit the activity of TGF β 1,so asto prevent the hepatic fibrosis.
