objectiveCholangiocarcinoma is a clinically common malignancy for which we lack effective therapy. Recently the role of XIAP(X - chromosome - linked inhibitor of apoptosis protein) in antiapoptosis has been greatly emphersized and effect on tumorigenesis promotion and prognosis. A lot of studies show that XIAP act as in inhibiting tumor cell apoptosis and in targets for therapeuty . XIAP was overex-pressed in cells of various cancers such as pancreatic cancer, ovariancancer and leukemias. Overexpression of XIAP may be a molecular marker of malignant transformation and progression . The overexpression of XIAP may play a key role in growth and metastasis of cholangiocarcinoma and serve as potential targets for therapeuty of cholangiocarcinoma. But there is few dependability research between the antigen and cholangiocarcinoma.In this study , we will examine the expression of XIAP in cholangiocarcinoma by immunohistochemistry(S -P). And by integrating clinical data,we will verify the expression of XIAP, and their relationship with clinicopathological significance of cholangiocarcinoma. We will analyze bionomics of cholangiocarcinoma in order to supply theoric bases for comprehensive treatment.Methods1. Tissue specimens collection: 41 cholangiocarcinoma specimens as experiment between May 1999 and October 2005 in the First Affiliated Hospital of China Medical University were obtained. Among 10 normal bile duct tissues ascontrol, ten is from transplant samples All the samples were confirmed by H&E before experiment.2. Main agents: XIAP antibody ( Affinity - purifiedGoat anti - human/ mouse/rat XIAP antibody ) are from R&D Systems,Inc. Peroxidase -conjugated AffiniPure Rabbit Anti -Goat IgG are from Santa Cruz Biotechnology,Inc. S- P kit are from Maixin Bio.3. Methods:The 41 cholangiocarcinoma specimens and 10 normal bile duct tissues are stained by immunohistochemistry ( S - P). and the expression of XIAP was clarified with microscope. Kruskal - Wallis test was used with SPSS11. 5 soft ware.Results1. The expression of XIAP in cholangiocarcinoma was higher than that in normal bile duct tissues, there was significant difference between them (X2 = 27.376, P<0.01)o2. Between male and female , there was no significant difference in the expression of XIAP( X =3. 250,P > 0. 05). Less than 60 ages and greater than or equal 60 ages, there was no significant difference in the expression of XIAP(X= 3.493 ,P >0.05). there was no significant difference in sex and age(P >0. 05).3. The expression of XIAP in well — differentiated, moderately differentiated and poorly differentiated cholangiocarcinoma was significant difference. The overexpression of XIAP in poorly differentiated cholangiocarcinoma there was significant difference between them(X2 =4. 729,P <0. 05).4. The expression of XIAP in defferent part of bile duct was no significant difference (X2 = 3. 133,P > 0.05 ).5. The expression of XIAP in lymphatic metastasis was significantly higher than that in non - metastasis (X2 =4. 066, P < 0. 05) , there was significant difference between them.Conclusion1. The overexpression of XIAP in cholangiocarcinoma .2. There was no relationship of the expression of XIAP in sex and age.3. The expression of XIAP in cholangiocarcinoma was correlated with differentiation and lymphatic metastasis.
