Effect Of Porphyra Polysaccharide On Mice Lymphocytes Secreting IL-2

PrefaceRecently, the study of marine drugs has become increasingly popular in the world. The chemistry and bioactivities of algae Polysaccharide are being researched futher in wide range. Porphyra is one kind of alge and Polysaccharide is one of the most components in Porphyra. A great many of studies both in home and aboard suggests that Porphyra Polysaccharide has various bioactivities, some of which have important immune - modulating effects. The purpose of this study is detect the level of IL - 2 which mice lymphocytes secrect by means of injecting Porphyra Polysaccharide into mice abdomen.Materials and MethodsExtracting polysaccharide from Porphyra yezoensis by take - downing the fat, protein and freeze - drying. 72 BALB/C mice were divided into 4 groups at random , Porphyra Polysaccharide in different dosages (0.05g · kg~-1, 0.25g · kg~-1 , 0. 5g · kg~-1 )were injected intraperitoneally into three of above groups. The left one was physiological saline solution alone as controls. Through 7 days injection, at 24h,48h,72h after the last injection,the level of IL -2 in the cultured supernatants of spleen cells from different groups of mice were measured by the method of ELISA. The data obtained were analyzed statistically by students t - test. All values were expressed as mean standard error of the mean ( S. E. M.) . Differences were considered significant at P<0.05 or P <0.01.ResultsThe levels of IL - 2 produced by spleen cells from the group of mice with the treatment of 0.25g ? kg"1 of Porphyra Polysaccharide were all showed to be significantly higher than those from other treatment groups. The levels of IL - 2 in the 0.05g ? kg"1 and 0. 5g ? kg"1 were not promoted statistically in contrast to the control group. At 24h after the last injection The level of IL - 2 begin to increase and reach to the peak at 48h,then declined to nomal by 72h.DiscussionPorphyra is a kind of alges which has high nutritional effects on animals. Porphyra yezoensis is the important component which possess various medical values, which have the functions antagonizing the hypercholesterolemia, phleg-masia, senility , tumor ,gene mutation and radiation obviously. Our study observed the levels of IL - 2 by injecting Porphyra yezoensis intraperitoneally. The result showed that a certain concentration(0. 25g ? kg"1) Porphyra yezoensis can enhance mice lympocytes secreting IL — 2, which has a curve relation with time. Previously research showed that Porphyra yezoensis can improve T cells proliferation, enhance the body immune function. IL -2 is mainly produced by activitied T cells, which support the conclusion of the study in theory. It' s mechanism is in studying.ConclusionPorphyra Polysaccharide of a certain concentration can improve IL - 2 secretion of mice and the secretion begain to increase at 24 h after the last immune injection, reach the peak at 48h,then declined to nomal by 72h. It suggests that PP has a improve effect tobody' s immune function.
