Objective To investigate the expression and the pattern of IGF-1R (Insulin-like Growth Factor-1 Receptor) in spinal cord and dorsal root ganglia after neurotomy of rat sciatic nerve .Methods Forty-five male adult Wistar rat s were divided randomly into control group ( n = 5)and experimental group ( n = 35).The rat s of experimental group were received a neurotomy to bilateral sciatic nerve and L4~L6 spinal cord and dorsal root ganglia were harvested respectively at 1d ,3d , 7d , 14d , 21 d , 28d postoperatively Immu -nohistochemical method and image analysis were used to study the expression of IGF-1R receptor 1 .Results IGF-1R were expressed in normal rat spinal cord and dorsal root ganglia in response to the neurotomy ,the expression of them reached a higher level, persisted for a short period of time and declined to normal level rapaidly, immunoreactivities of IGF-1R were also can be seen in glial cells and nerve fibers that localted in the white matter of spinal cordl .Conclusion The results reveal the favorable effect s of IGF-1 on nerve regeneration from the angles of central neurons ,which establishes the experimental and theoretic -calfoundation in the further treating peripheral nerve injuries with IGF-1.
