[Objective]Study on the expression of substance P to explore the effect of massage on the recovery of the mechanism of injury of sciatic nerve.(1)Through the thermal pain tolerance to observe the sensory response of rat.(2)Through the HE staining to observe the morphology of neurons in spinal cord and dorsal root ganglion.(3)Through the immunohistochemistry of the spinal cord and dorsal root ganglion in Substance P of quantitative analysis to observe the effects of massage.In these three aspects,from the mechanism of sensory pathway to further explain the massage in the treatment of peripheral nerve injury. To provide reliable basis for treating peripheral nerve injury diseases by clinical massage.To provide help for the development of massage subjects.[Methods]SD rats were selected as experimental animal, to prepare SNI model;Using random table to SD rats were divided into 5 groups:The normal group, the massage group, model group, sham operation group and the model control group.The massage group, the model group and model control group were prepared by SNI model. And the intervention of the massage group in 7 days after modeling.The control group was only given grasping bound model.Massage intervention:This experiment using the massage techniques simulator.Based on the SNI.In order to simulate the stiring method, kneading method, point method Massage intervention(acupoint selection, right of YinMeng, Chengshan, Yang Lingquan).(1) the thermal threshold of pain tolerance, observe the pain response in rats, the degree of recovery of sensory function.(2) to observe the morphology of neurons with HE staining, to evaluate the degree of neuronal damage.(3) by immunohistochemistry, Substance P, on the spinal cord dorsal root section in the quantitative analysis,to explore the mechanism of regeneration of peripheral nerve injury.[Results]1 Behavior-thermal pain tolerance experiments(1) 7 days after modeling,Pain reaction time of the model group was higher than that of the normal group had significant difference (P<0.05).Sham operation group was similar to the normal group (P>0.05).After SNI, the delay of pain response in rats.Because the pain pathway of obstacles. The nerve fibers arranged in disorder.The massage group:neuronal cells with mild degeneration and swelling, reduce the cell gap is not obvious, the most prominent nucleoli, small nerve fibers were scattered.Normal group and false operation group neurons look similar.The model control group and model group.(2) The HE staining was observed the level two of neuron in the spinal cord.Normal group:neurons were clearly visible nucleoli, nuclear center, arranged in neat rows, Nissl granules were clear. Model group:neuronal degeneration and swelling of the cells, reduce the number of permutations, scattered, and not see the Nissl body. The massage group:neuronal cells with mild injury, but most of the visible appearance of the normal, with regular arrangement, there are new neurons. Normal group and false operation group neurons look similar. The model group and the model control group, no significant difference (P>0.05).[Conclusion]1 Through the behaviorTo observe the result of thermal pain tolerance,Pain reaction time decreased,Massage can help the rat pain sense of heat recovery.2 By morphology,HE staining results,massage can protect neuronal cells and help SNI repair3 By histological and immunohistochemical method.The results of quantitative analysis of Substance P in spinal cord and dorsal root ganglion in the show,massage can promote the expression of Substance P in dorsal root ganglia of rat spinal cord.4 One of the mechanism of massage in the treatment of peripheral nerve injury Through the release of Substance P to control,Regulatory effect of Substance P on the neuroendocrine network is combined with the specific receptors and immune cells.Through the stimulation of Substance P receptors are the target cells release cytokines.The internal control environment, so as to protect the neurons to help repair damaged cells.The above,massage can help Substance P and the expression of pain and temperature sensation recovery.And the Substance P conduction and sensory nerve recovery had a positive correlation,The expression of Substance P can be used to evaluate the curative effect of massage and the degree of recovery of SNI.
