Objective To study the expression of Ki67, cysteine aspartate-specific protease-3 (Caspase-3) and E-cadherin(E-cad) in endometrial carcinoma and investigate the relation between Ki67, Caspase-3 and E-cad.Methods Expression of Ki67 , Caspase-3 and E-cad protein were detetected by immunohistochemistry method (EnVisionTM method) respectively in 15 cases of atypical hyperplasia of endometrium and 68 cases of endometrial adenocarcinoma and compared with clinical characteristics, such as age, clinical-stage and pathological grade.20 case of normal endometrium tissues were investigated as control group.Results 1. The expressing situation of Ki67, Caspase-3 and E-cad : The expression of Ki67 protein in endometrial adenocarcinoma was significantly higher than in normal endometrium tissues (P<0.01). The expression of Caspase-3 and E-cad protein in endometrial adenocarcinoma was significantly lower than in normal endometrium tissues (P<0.01).2. The relation between Ki67 expression and the clinical pathological characters of endometrial adenocarcinoma:The positive rate of Ki67 was strongly associated with the clinical stage (P=0.007) and pathological grade (P<0.01) . There was no relationship between Ki67 protein expression and other clinical pathological characters (P>0.05).3. There was no relationship between Caspase-3 protein expression and the clinical pathological characters in this experiment.(P>0.05).4. The relation between E-cad expression and the clinical pathological characters of endometrial adenocarcinoma : The positive rate of E-cad was strongly associated with the clinical stage. (P=0.018) , histological grade (P=0.006) ,muscle invasion (P=0.025) and lymph node matastasis (P=0.019) .5. The relation between Ki67,Caspase-3 and E-cad: The expression of Ki67 was negatively associated with Caspase-3 (r=-0.407 , P=0.001) .The expression of Ki67 was negatively associated with E-cad (r=-0.365 , P=0.002) .But the expression of Caspase-3 was positivelyly associated with E-cad (r=0.244 , P=0.044) .Conclusions 1.The expressing level of Ki67 in normal endometrium was lower than atypicalhyplasia and endometrial adenocarcinoma tissues . The expressing level of Caspase^3 in normal endometrium and atypical hyplasia was higher than in endometrial adenocarcinoma . The expression level of E-cad in normal endometrium was higher than in atypical hyplasia and endometrial adenocarcinoma tissues .All the results were highly significant. The results suggest that the expressing changes of Ki67, Caspase-3 and E-cad may activate the oncogene and play an important role in the happen of endometrial carcinoma.2.High expression of Ki67 and low expression of E-cad protein were related to clinical stage and pathological grade in endometrial carcinomas. The results show that high expression of Ki67 and low expression of E-cad may relate with the development of endometrial adenocarcinoma predict poor prognosis.3.The expression of Ki67 was negatively associated with Caspase-3 and E-cad, but there was a positive correlation beween Caspase-3 and E-cad.
