AIM: The ETS family of transcription factor is one of the biggest signal dependent transcription factor family, is involved in cellular proliferation, differentiation and malignance. The ETS family is highly conserved in evolutionary and shares a highly conserved DNA-binding regions which lies in C-terminal, was named ETS domain. ETS domain has a winged helix-turn-helix structural motif that recognizes and binds to a purine-rich DNA core elements of GGAA/T present in the promoter /enhancer regions of downstream target genes . Elf-1 is a member of the ETS family regulates genes involved in cellular development and differentiation , involved in developmental processes , mitogenic activation of cells, oncogenesis and viral gene activation and involved in the transcriptional regulation of several hematopocyte genes. Enhanced expression of Elf-1 has been reported in prostate cancer , endometrial carcinoma and osteosarcoma. MMP-2 is one of the matrix metalloproteinases, is an important proteolysis in tumor's invasion and metastasis. To study the expression of E74-like factor 1 (Elf-1), matrix metalloproteinases-2 (MMP-2) in human breast carcinoma and to explore the role of Elf-1 in the angiogenesis and metastasis of carcinoma.METHODS: The expression of Elf-1 and MMP-2 protein were detected in paraffin section tissues of breast carcinoma(n =60),lobular hyperplasia of the breast (n = 13), fibroadenoma (n = 15) and normal breast tissues beside carcinoma (n = 7) by the immunohistochemical SP method respectively. We analyzed the relationship between Elf-1 protein expression and characteristics of patients with breast carcinoma: tumor's size, clinical stage, histological grade , pathologic type as well as lymphatic metastasis. RESULTS: Elf-1, MMP-2 were negatively expressed in normal breast tissues beside carcinoma, The positive rates of Elf-1, MMP-2 expression were 3 (23.1%), 3(20%) and 4 ( 30.8% ), 3 (20% ) in lobular hyperplasia of the breast and fibroadenoma respectively. The positive rates of Elf-1, MMP-2 expression were 50 ( 83.3% ) 和 47 ( 78.3% )in breast carcinoma and significantly higher than normal breast tissues beside carcinoma (P < 0.0001;P < 0.0001 = , lobular hyperplasia of the breast (x~2=16.6, P <0.01;x~2=9.33, P<0.01= and fibroadenoma (x~2=20.3, P<0.01;x~2= 18.375, P< 0.01 =The expression of Elf-1, MMP-2 were significantly correlated with tumor's size (x~2=3.923, P < 0.05;x~2=5.28, P<0.05 ), histological grade( x~2=3.92, P< 0.05;x~2 =8.59,/J< 0.01 ) , TNM stage (.v:=4.36, /J<0.05;.v2=5.88, P < 0.05 ), pathologic type (.?"= I0.78,P<0.01;a 2=631,P < 0.05 ) and lymphatic metastasis (x 2=3.92, P<0.05;.v 2= 6.37,P < 0.05). Elf-1 expression was positively associated with MMP-2 level (/? =0.525, P <0.0l).CONCLUSION: Elf-1 was over-expressed in breast carcinoma, and its expression was related to tumor's size, clinical stage, histological grade, pathologic type and lymphatic metastasis. Elf-1 expression was also positively related to MMP-2 level.
