Objective: To detect the mutation frequency of hmsh2 and hmlhl gene in lead poisoning placenta with mouse, investigate the relationship between the mutation of hmsh2, hmlhl gene and the pregnant maza with mouse, in orde to further explore the molecrlar genetic mechanism.Methods: Genome DNA of 17(9) samples with nonpregnant mouse placenta after lead intoxication;22(9) samples with progestation mouse placenta after lead intoxication;25(9) samples with duration of pregnancy mouse placenta after lead intoxication, and 15 normal samples as the control,subsequently the exonl2 of hmsh2 (hmlhl) was amplified with PCR. The result was analyzed with SSCPoResults: â‘ Mutations of the exonl2 of hmsh2 were identified as following :5/17 in the nonpregnant mouses;8/22 in the pregnancy mousse,and 10/25 in the duration of pregnancy mouses while there are no Mutations in normal samples been found. â‘¡Mutations of the exonl2 of hmlhl were identified as 5/9 in the nonpregnant mouses;4/9 in the pregnancy mousse,and 6/9 in the duration of pregnancy mouses while there are no Mutations in normal samples. Results show: There were no obvious difference in the mutation rate of several Stagings (P>0.05),but significant difference have been found between the contral and the lead intoxicated sampals. (P<0.05);â‘¢ The mutation rate of hmsh2 and hmlhl gene in the plumbism mouse was significantly higher than that of normal mouse.(P<0.05).Conclusions: As a result: â‘  There were no obvious difference in the mutation rate of several Stagings (P>0.05) with the plumbism mousse both the hmsh2 and hmlhl gene,but significant difference have been revealed between the contrOl and the lead intoxicated sampals. Which indicate that Plumbi may contribute to the mutation of hmsh2 and hmlhl gene with pregnant mouses. Maybe there is a closely relation both them.
