| Objective: The objective of this experiment is to explore the relationship between β-catenin , cyclin D1 and HCC so as to show the mechanism of pathologic transformation of liver cancer cells. Thus, theoretical proof can be provided for the prognostic evaluation of HCC and the intervention of clinic prophase and possible approaches can be found to stop the spread of HCC.Method: Pathological team selected 48 HCC samples that are removed during operation and are confirmed by the Department of Pathology. Immune-histochemical staining of β-catenin ,cyclin D1, HBV exterior antigen and HCV antigen mono-clone antibody were detected with S-P (Streptavidin-Peroxidase)method. Statistical analysis was executed by SPSS 10.0 software.Results: β-catenin of normal liver sample is cell membrane positive with no visible positive signals of cell plasma and nucleus. There's no excessive display of cyclin D1 on cells. The percentage of abnormal display of β-catenin in HCC tissue is 72.9% ( 35/48 ) , the percentage of display of the weakened cell membrane is 52.8% (25/48) and the percentage of display of abnormal position of plasma and nucleus is 66.7% (32/48). β-catenin in the cancerous tissue shows consistent coloration of cell membrane and there's no visible positive signal for plasma and nucleus, although a small portion of the sections show weakened cell membrane with display percentage of 8.3% ( 4/48 ) . Hence there's remarkable difference (X~2 = 32.4, P < 0.01) . The weakened display of β-catenin cell membrane is irrelevant with gender of HCC patients, AFP, and the diameter of the tumor. It is relevant with cancer peg and envelope and the phase of the tumor. The display of abnormal position of β-catenin is irrelevant with gender of HCC patients, AFP, cancer peg and envelope, but it is relevant with the diameter and phase of the tumor. There's no excessive display of cyclin D1 in cancerous tissues, but the display percentage in liver cancer tissues is 45.8% (22/48) . There's significant difference (X~2 = 26, P < 0.01 ). The display of abnormal position of cyclin D1 is irrelevant with gender of HCC patients, AFP, cancer peg and envelope, but it is relevant with the diameter and phase of the tumor. There's no significant difference between the display of the weakened β-catenin cell membrane and the excessive display of cyclin D1 (X~2 = 0.10, P > 0.2) while there's significant difference between the display of abnormal position of β-catenin and the excessive display of cyclin D1 (X~2 = 7.09, P< 0.01) . The display of weaked β-catenin cell membrane and the excessive display of cyclin have no relevance with the positive display of HBsAg , HCV, yet the display of abnormal position of β-catenin is relevant with the positive display of HBsAg, HCV and there's significant difference ( U= 2.0, 0.01 < P < 0.05 ) .Conclusion: 1. β-catenin has abnormal position and weakened cell membrane display in the HCC tissues. It may participates in the emergence and development of HCC.2. The abnormal display of β-catenin is relevant with HCC pathological classification and cancer peg and envelope. It may be of used as a reference to estimate the degree of the malignance HCC and the potential of the attack.3. The excessive display of cyclin Dl is relevant with HCC pathological classification. Cyclin Dl has close relationship with the development of liver cancer and can show the impact of liver cancer.4. The excessive display of β-catenin is relevant with the excessive display of cyclin Dl. There may be cooperated function of the two during the development of HCC.5. The excessive display of β-catenin is relevant with the infection of HBV and HCV. The gateway of Wnt signal transmission may be one of the approaches that virus infection leads to cancer. |
