Objective: Hepatocellular carcinoma (HCC) is one of the common types of malignant tumors with poor prognosis, metastasis and high fatality rate. Traditional Chinese Medicine (TCM) has been describing and treating cancer with varying success for millennia. TCM can improve the life quality, delay the cancer development, recrudescence and metastasis. It is recently demonstrated that TCM inhibits proliferation of tumor cells and triges apoptosis. As in many other types of cancer, aberrant activation of the canonical Wnt/beta-catenin signaling pathway is an important contributor to tumorigenesis. Several laboratories have shown that the Wnt/beta-catenin pathway plays an essential role in all phases of liver development and maturation, and is required for the metabolic function of this organ. In this paper, we explore the expression of beta Catenin and Cyclin D1 in the hepatocarcinoma 22 tumour-bearing mice and the effect of HuaJiYiGanJian(HJYGJ).Methods: 60 mice models were built by hepatocarcinoma H22 tumour with an indirect method. After 24h of postoperation, the 60 mice were distributed randomly into 4 groups: each group had 15 mice. Control group were treated by intragastric administration with normal saline for 15d, qd. TCM group were treated by intragastric administration with HJYGJ(complex prescription of Chinese crude drug)for 15d, qd. Western Medicine group were treated by intraperitoneal injection with cylophosphamide for 15d, qd. TCM+Western Medicine group were treated by intragastric administration with HJYGJ and intraperitoneal injection with cylophosphamide for 15d, qd. The therapy effect was observed and the tumor inhibitory rate was calcu- lated after killing the animals on the 15th day after administering and beta Catenin and Cyclin D1 was detected by immunohistochemistry.Results:â‘ the tumor inhibitory rate of TCM group was 38.35%, the tumor inhibitory rate of Western Medicine group was 55.48%, the tumor inhibitory rate of TCM+Western Medicine group was 70.93%, the difference is significant(Kraskal-Wallis Test P<0.05). The tumor inhibitory rate of TCM+Western Medicine group (70.93% )was higher than in Western Medicine group (55.48%), the difference is significant(Mann-Whitney Test P<0.05). TCM could elevate antitumor activity with cyclophosphamide.â‘¡The result of immunohistochemistry showed that the expression rate of beta Catenin in control group, TCM group, Western Medicine group and TCM+Western Medicine group was 85.71%, 78.57%, 71.43% and 30.77% respectively, the difference is significant(P<0.05). There was decreased expression of beta Catenin after the treatment. The expression rate of Cyclin D1 in each group was 92.86%, 75.87%, 64.29% and 23.09% respectively, the difference is significant(P<0.05). There was decreased expression of Cyclin D1 after the treatment.Conclusion:â‘ HJYGJ is inhibitive to hepatocarcinoma H22 and syner- gic with CYC.â‘¡It inhibits the proliferation, invasion and metastasis of hepatocarcinoma H22 by affecting the Wnt signal-pathway.
