ObjectiveTo study the anti-tumor effects induced by 2, 3-dioxoindoline in SH-SY5Y cells. MethodsAccording to the final concentration of 2, 3-dioxoindoline,the SH-SY5Y cells were divided into 5 groups:â‘ 2, 3-dioxoindoline 50umol/L group â‘¡100umol/L group â‘¢200umol/L group â‘£500umol/L group (5)the control group. There is an explaniation that MTT assay was done in the five groups that are stated above,but the other experiments only had the four groups except the first group.MTT assay was used to examine the effect of 2, 3-dioxoindoline on the proliferation of SH-SY5Y cells;We observed and evaluated the survival of the cells after and before the administration of 2, 3-dioxoindoline.Flow cytometry was used to determine cell cycle and cell apoptosis.The mRNA levels of bcl-2, bax and vegf were detected by RT-PCR.The activity of caspase-3 in SH-SY5Y cells was assayed by immunohistochemical technology.The statistics of variables we have achieved were treated with SPSS 12.0.Results2, 3-dioxoindoline inhibited the proliferation of SH-SY5Y cells and induced cell apoptosis.The OD value of MTT reduction assay showed that 2, 3-dioxoindoline markedly inhibited the growth of SH-SY5Y cells separately while the control group showed no effects on SH-SY5Y cells.Comparing with the control group,the 2, 3-dioxoindoline group had statistical significance.At the same time it also affected the distribution of cell cycle,that is to say,it increased cell population in G0/G1 phase and decreased it in S phase.The S phase is a period DNA synthesizs;The conseqence proved that 2, 3-dioxoindoline could prompt the failure in the synthesis of DNA in tumor cells.RT-PCR data showed that bcl-2 mRNA and VEGFmRNA level were markedly downregulated in SH-SY5Y cells administered with 2, 3-dioxoindoline for 48 hours,while the control group showed no effects on SH-SY5Y cells.What we have to stress is that the decrease of bcl-2 mRNA in SH-SY5Y cells was in a concentration dependence manner.And the level of bax mRNA kept constant during the treatment.The immunohistochemical technology showed that the caspase-3 protein in SH-SY5Y cells rised after adiministration of the drug. Conclusions2, 3-dioxoindoline inhibits proliferation of SH-SY5Y cells with induction of cell apoptosis and cell arrest.The mechanism of apoptosis is possibly related with down-regulation of bcl-2 gene expression,up-regulation of caspase-3 protein,the change of cell cycle and prevents vascular-forming and the transmisson of signals in tumor cells.In conclusion,2, 3-dioxoindoline may be a more promising and effective drug against tumors.
