| Construction and package of the expression plasmid pAdEasy-1 System encoding the human papillomavirus 16 E7 and E7's influence to the cell cycleAbstract: Objective: The study was designed to investigate the effect of the human papillomavirus 16 E7 gene on cell cycle ,constructing and expressing human papillomavirus 16 E7 gene with adenovirus vector.Methods: Constructed and packed recombinant adenovirus which expressing E7 gene.Flow cytometry was used to detect the changes of cell cycle-phase and cyclinDl between cells infected and uninfected by recombinant adenovirus.MTT was used in the detection of the alteration of cell increment.Result: The recombinant adenovirus had stable efficiency of infection and E7 genes could be expressed stably.MTT showed the cell multiplication of Hela cells was accelebrated after infected. FCM showed cells in S phase and M phase increased obviously than uninfected.Conclusion.The recombinant adenovirus expressing E7 gene was able to infect target cells.E7 gene can made cell cycle confused which can be assumed to restrain and heal cervical cancer. |
