| Object: To study whether the n-butanol extract from Lagotis Brevituba Maxim extracted might inhibit Human Stomach Cancer SGC-7901 cells proliferation or induce apoptosis. Research the immunoregulation activities of the extract from Lagotis Brevituba Maxim. The discoveries provide an important basis for possible medical application.Methods: Human Stomach Cancer SGC-7901 cells were treated with the n-butanol extract from Lagotis Brevituba Maxim at different concentrations for 24h~ 72h. Cell proliferation was evaluated by MTT assy. Morphological changes of apoptosis were examined by fluorescence microscope and electron microscope. DNA fragmentation was visualized by agarose gel electrophoresis. The amount of apoptosis cells was measured by flow cytometry. Mice with sarcoma-180 were fell into five groups: NS group , CY (20mg/Kg·d)group, the n-butanol extract from LagotisBrevituba Maxim (500 mg/kg·d, 1000 mg/kg·d, 1500 mg/kg·d) groups. The control group was built at the same time . Through observing the weights and the tumoralleosis of mice, measure anti-tumor rate and take pictures;Proliferations of T lymphocyte were measured by MTT method;The function of B lymphocyte was measured by antibody agglutination potency of serum against SRBC;The consistence of IL-2> TNF-a were measured with radioimmunoassay.Results:1. Growth of SGC-7901 cells was obviously inhibited by the n-butanol extract from Lagotis Brevituba Maxima? at concentrations from 0.34 mg/ml to 21.60 mg/ml. The anti-tumor activity was gradually enhanced with Lagotis Brevituba Maxim variety of time-and-dose.2. After incubation of SGC-7901 cells with the n-butanol extract from Lagotis Brevituba Maxima* at concentration of 2.7mg/ml for 48h, morphological changes of apoptosis were observed. DNA ladder was observed by DNA gel electrophoresis.3. The n-butanol extract from Lagotis Brevituba Maxim can repress tumor growth at concentration of 1000mg/kgd and 1500mg/kgd (p<0.01) . Anti-tumor rates are 37% and 48% respectively.4. The proliferation of T cell was strengthen by the n-butanol extract from Lagotis Brevituba Maxim when PHA existed ( P<0.05 ) .5. The contains of IL-2 and TNF-a in serum of mice with sarcoma-180 were increased by the n-butanol extract from Lagotis Brevituba Maxim (1000~1500mg/kg-d) (P<0.05) .6. Antibody agglutination potency of serum against SRBC was elevated by the n-butanol extract from Lagotis Brevituba Maxim at concentrations of 1000 ~ 1500mg/kgd (P<0.05) . Conclusions:1. The growth of SGC-7901cell was inhibited by the n-butanol extract from Lagotis Brevituba Maxim ,and apoptosis of SGC-7901cell was induced, too.2. The growth of tumor cell in mice with sarcoma-180 was repressed by the n-butanol extract from Lagotis Brevituba Maxim.3. The functions of T and B lymphocyte were enhanced by the n-butanol extract from Lagotis Brevituba Maxim. |
