Lagotis brevituba Maxim(L.brevituba Maxim)belongs to family of Scrophulariaceae and the Lagotis genus,which mainly distributes in Tibet,Qinghai and Gansu.For a long time in the past,it has been used as a Tibetan medicine to treat fever,nephritis,lung disease and other diseases that in reference to antioxidant capacity.Up to now,phenylethanoid glycosides and phenols were the main chemical constituents that have been identified in this plant,and more chemical constituents of it need to be explored.Because many diseases are closely related to antioxidant activity,the chemical composition and antioxidant activity in vivo and in vitro were studied in this paper.Incarvillea compacta Maxim is a plant that of the genus Incarvillea and the famiy of Bignoniaceae.It is often found in Lhasa and Changdu of Tibet,Yushu and Haixi of Qinghai,Batang and Muli of Sichuan,etc..It taste bitter and has the functions of smoothing circulation,relieving pain and regulating menstruation and hemostasis.At present,about 5 literatures have studied the chemical constituents of Incarvillea compacta Maxim,in-cluding flavonoids,phenylethanoid glycosides,terpenoids and volatile components.In terms of pharmacological activity,its anti-tumor and hepatoprotective effects have been explored.However the analgesic activity,which reflected in other plants of the same genus,has not been reported.Therefore,in order to make these traditional Chinese medicines with regional characteristics widely used,it is necessary to explore more information about their chemical composition and pharmacological activities.1.Studies on the Chemical Constituents and Antioxidant Activities in Vitro and in Vitro of Lagotis brevituba MaximIn this section,the chemical constituents of Lagotis brevituba Maxim were characterized,and the contents of total flavonoids,total phenols and total saponins were determined.Free radical(DPPH,ABTS)scavenging activity test,reduce power test,?-carotene/linoleic acid cooxidation assay and mouse model of radiation injury were used to evaluate their antioxidant activities in vivo and in vitro.The results showed that the contents of total flavonoids,total phenols and total saponins in water extract were the highest.A total of 44 compounds were identified,including 19 flavonoids,14 phenols,8 phenylethanoid glycosides,2 iridoid glycosides,and compounds 1-25 were identified for the first time from the genus Lagotis.In DPPH,ABTS and reduce power tests,water extracts have the best antioxidant ability.The methanol extract showed the best antioxidant activity in the ?-carotene/linoleic acid cooxidation test,probably because the polarity of methanol was closer to the oil system in this experiment.In the mouse model exposed to radiation,compared with no treatment group,the extract of Lagotis brevituba Maxim could increase the levels of SOD and GSH-PX in a dose-dependent manner,and also decrease the contents of MDA,so that the indexes of mice tended to be normal.The results of this section can provide reference for the future research of Lagotis brevituba Maxim and also indicate that it can be used as a potential plant source of antioxidants.2.Chemical Constituents?Analgesic and Antioxidant Activities of Incarvillea compacta MaximIn this section,the content of total phenols,total flavonoids and total alkaloids in different extracts of Incarvillea compacta Maxim was qualitatively analyzed by HPLC-MS.At the same time,the antioxidant and analgesic activities of Incarvillea compacta Maxim were also studied.The results showed that the content of various components in methanol extract was the highest.Twenty-five components from Incarvillea compacta Maxim were analyzed,of which compounds 1-23 were firstly identified in this plant.Compared with methanol extracts with more flavonoids,phenols and alkaloids,water extracts have the best ability to inhibit oxidation.Therefore,it is infered that other ingredients in this plant exert a considerable function on antioxidant activities.Also,the water extract elicited dose-dependent analgesic effects in the first and second phase in a formalin test,and augmented the latency of pain from a hot-plate test.The water extract decreased the amount of writhing compared with the control group in the acetic acid-induced writhing test significantly.The results showed that Incarvillea compacta Maxim could be used as a plant source of new antioxidant and analgesic.The data obtained in this study could strengthen its application in the medical industry and supply its further study with reference.
