Tumor is organism under the function of any kind ofcarcinogenic factor, the partial tissue cells lost its normal growthregulation in the gene level, causes hyperplasia and then form theneoplasma. The multiplication ability enhance and apoptosis inhibitin cells are two main mechanisms which lead to tumor occurs.Cellapoptosis under gene control is playing the vital role indifferentiation, growth and maintaining normal internal environmentstably.The inhibition of cell apoptosis and the occurrence ,development of the malignant tumor as well as chemotherapy andradiotherapy insensitively have close relation.Survivin has recently been identified as a new member of thetumor of the inhibitor of apoptosis protein (IAP) gene family. It hasmany kinds of biological function, such as inhibiting the activenessof active proteinase caspase, participating cells cycle regulation andadjusting the cell segmentation and so on. It has close correlationwith the occurrence, development of the tumor.At present studiesindicated surviving gene expresses in the most malignant tumor, ithas obvious differences among the degree, spot, the way of theexpression and the corresponding tumor architecture. The relationsof survivin gene expression and clinical stage, the tumor architecturegrade, the pathology type, the metastasis and the erosion of thecervical cancer were indefinite, the conclusion was inconsistent andwait for further studies.The cervical cancer is also called invasive carcinoma of cervixuteri. It is a common gynecology malignant tumor whose incidenceis only inferior to the breast cancer in global women. It seriouslythreatens women's health and their life. Cervical cancer takessquamous cell carcinoma as familiar, occupies 80%-85%. Thecause of disease in cervical cancer is not known distinctly now, maysynthesized by various factors.Predicting the erosion and metastasis ability of cervical cancerhas the vital significance to understand the pathological change andthe biological characteristic of the disease, formulates the correcttreatment plan and judge prognosis .Unfortunately, we have not yetdiscovered the specific biological index at present which caneffectively predict the erosion and the metastasis ability of cervicalcancer.To explore the relationship between the expression of survivingene and the occurrence and development of cervical cancer,approach the clinical significance,immunohistochemical PV methodwas used in this experiment to examine the expression of survivinggene in 54 cervical cancer tissues and 16 normal cervical tissuefrom which uterus were ectomized because of hysteromyoma ,analyzed its clinical index , is for the purpose of studying the ruleand the clinical significance of survivin gene in cervical cancer .Materials and methods54 cases of cervical squamous cell carcinoma who were treatedand combined surgical operation in our hospital from January,2003to April,2005 were selected .They were divided into two groups,young group ,below 35 years old of 26 cases(average 30 years old) ,middle and old aged group , over 45 years old of 28 cases (average55.18 years old).Marked a comparison between two groups ofcervical squamous cell carcinoma patients in clinical stage, thepathology graduation and the lymph node metastasis, there are nostatistical significance (P > 0.05). Stochastic selected 16 cases fromwhich uterus were ectominzed because of leiomyoma-uteri in thesame time to take the comparison.Specimen origin: The wax blocks were stock in the pathologydepartertment of our hospital. 54 specimen of cervical squamous cellcarcinoma were all null-radiotherapy and null-chemotherapy.Using immunohistochemical method, I examined theexpression of survivin gene in 54 cases of cervical squamous cellcarcinoma and 16 normal cervical tissues and analyzed therelationship of the clinical characteristics and pathological features.Results(1)Survivin-positive staining mainly located in the cytoplasm,for different sizes of yellow brown particles, and some of themwere covered distribution diffusely.(2)The expression of survivingene was not detected in normal cervical tissue . In contrast,surviving gene was expressed in 37 of 54(68.5%) cases of cervicalsquamous cell carcinoma.(3)The positive rate of survivin gene inyoung group of cervical squamous cell carcinoma was significantlyhigher than middle and old aged group (p<0.05=. (4) The laterclinical stages of the patients ,the higher rate of survivin-positiveexpression ,the difference has statistical significance(p<0.05=.(5)The positive rate of survivin gene in the group of lymph nodemetastased was significantly higher than lymph nodenull-metastased (p<0.05). (6) There is no relationship betweensurvivin-positive expression and the pathology grades.Conclusion(1) Survivin gene was not expressed in the normal cervicaltissue, but high expressed in cervical squamous cell carcinoma.Survivin play certain role in the occurrence of the cervical cancer. Ithas great value to diagnose the cervical squamous cell carcinoma.(2) There were relationships between survivin gene expressionand age, clinical stages, lymph node metastasis. The patient isyounger or the clinical stage is later, the survivin gene expression ishigher. The survivin gene expression is significantly higher in thegroup of lymph node metastasized than lymph node null-meta-stasized.The stable expression of survivin in cervical squamous cellcarcinoma indicates that the tumor has high invasion and poorprognosis. This will reflect the malignant degree in cervicalsquamous cell carcinoma. The expression of Survivin gene maybecome the effective index to forecast the invasion of cervicalsquamous cell carcinoma, judge prognosis, and instruct the clinicaltreatment.
