| Background: Cervical carcinoma is the most frquently seen malignanttumor of female reproductive system..It has a high morbility and tends to becaught by more young women.The tumorigeness is often related to the derangement of the cell cycle.Skp2 is a member of F-box protein. Many researches show that Skp2 caninteract with many cell cycle regulating factors including p27, and lead to theirubiquitination and degeneration. So it is very important in DNA duplication,regulation of transcription and cell proliferation. p27 is a kind of cell dependantkinase inhibitor, and negtively regulate the cell cycle. Many researches provedthat p27 is an anti-oncogene, and the over-expression of Skp2 often lead to thedegeneration of p27, cause the derangement of the cell cycle and result in thetumorigenesis. In recent research, Skp2 is related with drug resistance andinticated poor prognosis in a majority of tumors. However, there is still fewreport about protein expression of Skp2 in Cervical carcinomaObjective: To investigate the protein expression of Skp2 incervical carcinoma and cervical intraepithelial neoplasia(CIN) and see therelationship between Skp2 and canceration of cervix.Method: The experimential object is devided into three groups includingcervical carcinoma , cervical intraepithelial neoplasia and normal cervixtissue.And the protein expression of Skp2 is estimated by S-Pimnunohistochemistrial technique.Result:(1)The protein of Skp2 is in lower level expressed in normal cervixtissue;The protein of Skp2 is highly expressed in cervical carcinoma andcervical intraepithelial neoplasia and the stain intensity is deep;The expressionrate of Skp2 in cervical carcinoma and normal cervix tissue Was significantlydiferent(p<0.05)。(2) In cervical intraepithelial neoplasia, the expression rate of Skp2 wassignificantly diferent(p<0.05) comparing to the expression rate in normal cervixtissue.(3) In cervical carcinoma, there is no significant diference(p>0.05) ofexpression rate of Skp2 in diferent pathologic type,stage or age.Conclusion: (1) The intensity of Skp2 expression is high incervical carcinoma, Skp2 might play an important role in the development andprogression of cervical carcinoma.(2) The high expression of Skp2 may be correlated with the malignancy ofcervical carcinoma . |
