Objective To evaluate the difference of mitral valve replacement under Beating heart and mitral valve replacement(MVR) arrested heart with moderate hypothermia by detecting the expression of heat shock protein 70(HSP70) in the myocardial tissue and level of antibody against HSP70 in serum of MVR patients.Methods 32 patients with defect of mitral valve attacked by rheum heart disease were divided into observation group(under Beating heart) and control group (arrested heart with moderate hypothermia 25℃~28℃) randomly. The expression levels of HSP70 in myocardial tissue in 17 MVR patients under beating heart and 15 controls patients were detected by immunohistochemical staining and analyzed by computer image-pattern analysis system. The serum anti-HSP70 antibody levels in cases and controls were measured by enzyme-linked immunosorbent assay (ELISA).Results (1) The hematoxylin and eosin-stained sections of myocardium samples showed that the hypertrophy of the ventricular myocytes and interstitial swelling were evident in observation group compared to those in the control group .The expression of HSP70 increased significantly in the hearts compared to the control hearts (P<0.05) . Most of the HSP70 were observed in cytosol other than in nucleus or membrane . the average optical density (AOD) of HSP70 expression in the observation group was 0.39±0.08, and it was noticeably increased as compared to that of the control group (0.22±0.06), P<0.05...
