| Objective:to assess the effect of esomeprazole-based triple therapy on healing of duodenal ulcer with Helicobacter pylori infection. Methods: Randomized , single-blind,paralled-controlled study was conducted at the first Affiliated Hospital of Guangxi Medical University during the period of May to December in 2005. 66 patients with H.pylori infection proven by both histology and rapid urease test and active duodenal ulcer were divided into three groups: A ,B and C. All patients were given esomeprazole-based triple therapy for the first week: esomeprazole 20mg bid , amoxicillin 1 g bid and clarithromycin 500mg bid. Then the patients in group B were given esomeprazole 20mg qd for another week. Whereas the patients in group C were given esomeprazole 20mg qd for three weeks. The eradication rate of Helicobacter pylori and the healing rate of duodenal ulcer were evaluated by the second endoscopy on the 28tth day after completion of treatment . The relief of the symptoms of epigastric pain, sour regurgitation and heart-burn were also evaluated on the 28th day. Results: 58 patients completed the trial. 8 patients dropped out. The eradication rates of H.pylori in groups A, B, C were 88.88 %,90.47 %,and 94.73 % respectively, there were no significant difference among the three groups(P>0.05). The ulcer healing rates in groups A, B, C were 83.33 %,85.71 % and 89.43 %; respectively, there were no significant difference among the three groups(P>0.05). The total score of symptom relief in group A , B, C ( x±1.96 s) were 11.66±4.86; 13.50±7.53;16.58±10.27, the difference was significant (P<0.05). Conclusions: Three regiments can heal the ulcers and eradicate H.pylori effectively, the symptoms of these patients were improved at the same time. Furthermore the degree of symptom relief was related to the time of esomeprazole being used.
