Isolation Alkalioids From Sophora Flavescens Ait And Study On Their Anti-tumor Activities.

"Kushen",the dried roots of Sophora flavescens Ait, is the commonly used traditional Chinese medicines derived from Sophora plants.It's Chemical constituents contain quinolizidine alkaloids, prenylated flavonoids and oleanene triterpenoids.Recently, there are increasing study on the alkaloids's inhibitory effects on tumor cell lines and more attention has been paid to its anti-tumor effects both in vitro and vivo.In order to develop new anti-cancer pharmaceuticals from Chinese herbs medicine and explore new anti-tumor mechanisms ,four alkaloids were isolated from the ethanol extracts of the roots of Sophora flavescens Ait by repeated silicagel column chromatography .The structures of four compounds were identified by IR,MS,1H-NMR,13C-NMR methods as sophocarpirine,matrine, sophoddine and oxymatrine.To investigate the anti-tumor activity of four alkaloids on ovarian cancer cell line SKOV3 and human lymph mode high metastasis ovrian carcinoma cell line (PM-2). MTT assay, Immunohistochemistry, Hematoxylin-Eosin staining and Transwell Migration Assays are used. the MTT results show that the inhibitory effects of four alkalinids on SKOV3 and PM-2 cell are lower than Oxaliplatin ,but sophocarpirine's inhibitory is the best one among the four alkalinids.Under the light microscope , After treatment with four alkalinids, SKOV3 and PM-2 cells showed typically morphologic features changes of cell crimple,nucleus shrink, nucleus border gather and have a great deal air bladder in their plasm. The expression of VEGF and MMP-2 were remarkably surpressed in SKOV3 and PM-2 cells after treatment with four alkalinids.
