Background Cardiovascular disease caused by atherosclerosis is a common disease damaging people,s health. It is an important topic in medicine to study the mechanism of atherosclerosis and its prevention and therapy. Ross thought atherosclerosis is a chronic hyperplasia inflammatory caused by injury factors. Intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) is a critical adhesion molecule existing on vascular endothelial cells, which promotes leukocytes adhering to endothelial cells. The non-lipid effects of statins such as anti-infammotory, stabilizing plaque ect are increasingly recognized.Aim To observe the influence of atorvastatin (ATV), a kind of 3-Hydroxy-3-methylglutary coenzyme A (HMG COA) reductase inhibitors (statins), at different concentrations on the mRNA expression of ICAM-1 and VCAM-1 in cultured human umbilical vein endothelial cells (HUVEC) activated by tumor necrosis factor-α(TNF-α) in order to understand the non-lowing lipid of atorvastatin.First Part The expression of ICAM-1 and VCAM-1 mRNA in HUVECs activated by TNF-αMethodsHUVEC were isolated from freshly obtained human umbilical cords by collagenase type II treatment. The cells were cultured in M1640 supplemented with 10% bovine colf serum and passages 2 to 3 were used in this study . HUVECs were coincubated with different concentrations (0μg/L ,10μg/L,20μg/L,30μg/L, 40μg/L) of TNF-αfor 48h or with 30μg/L TNF-αfor different periods (0, 6, 12, 24, 48, 72h). ICAM-1 and VCAM-1 mRNA was detected by quantitative reverse transcription polymerase chain reaction (RT-PCR). The protein of ICAM-1 and VCAM-1 was detected by Western blotting.ResultsThe levels of mRNA of ICAM-1 and VCAM-1 increased initially after cells were coincubated with 10μg/L TNF-α(P<0.01), and reached peak value at 30μg/L, and then declined; In the same concentration of 30μg/L TNF-α. the expression of mRNA of ICAM-1 and VCAM-1 were increased significantly with the coincubation time from 0h to 48h (P<0.01), and maintained high level to 72h.Second Part The inhibition effect of atorvastatin on the mRNA Expression of ICAM-1...
