Objective: To explore the effect and mechanism of benzo[a]pyrene on central nervous system function in coke oven workers and rats.Methods: 1. Health adult male coke oven workers were selected from a coke plant of one state-owed enterprise in Taiyuan City, and were classified into three exposure categories according to their work sites: bottom-oven group, side-oven group and top-oven group. The controls were selected from the native material plant of the same enterprise.â‘ Concentration of B[a]P in the work environment was monitored.â‘¡Blood and urine samples were collected immediately after a shift at the end of a working day. The level of urinary 1-hydroxypyrene(1-OhP) was measured by High Performance Liquid Chromatography (HPLC). Levels of antioxidase and lipid peroxidation(LPO)were detected by test kits.â‘¢The workers'general information correlated with the investigation was collected by query and filled in the questionnaire by special investigators who were specially trained according to the same criteria.â‘£WHO recommended NCTB were performed on coke oven workers and controls to test the neurobehavioral changes and the mood state.2. Forty male SD rats, adult and healthy, were divided into 4 groups at random: normal oil control, B[a]P1.3mg/kg,B[a]P3.2mg/kg,B[a]P7.8mg/kg. They were exposed to B[a]P by intraperitioneal injection for 60d. After exposure,â‘ The learning and memory function was measured by Morris water maze.â‘¡The apoptosis of neurocyte in rats'hippocampus was measured by flow cytometry.â‘¢The morphology of neurocyte in rats'hippocampus was observed by light microscope and transmission electron microscopeâ‘£Enzymes in blood(SOD,GSH) and mitochondria (SOD, SDH, MAO, AChE,Na+-K+ ATP, Ca2+-Mg2+ATPase)and levels of MDA,ROS in blood were detected by test Kits. Results: 1â‘ No significant differences were found among four groups in age, length of service, smoking and drinking. However, there was significant difference in education; There were no significant differences in the scores of the symptomes of digestive system(SDS) and cardiovascular system(CVS) (P>0.05).However, there were significant differences in the scores of the symptomes of nervous system(SNS), the symptomes of endocrine system(SES) and the symptomes of skeletal musculature...
