Objective: To study the expression of metastasis suppressor gene KAI1,high-risk human papillomavirus type16/18E6 and E7 in human cervical tumors and to investigate their possible roles in tumor formation and progression. Methods: To detect the expression of metastasis suppressor gene KAI1, human papillomavirus type 16/18 E6 and E7,117 paraffin embedded samples of different cervical tumor were examined using immunohistochemical staining technique, including control group were composed of 20 cases of normal cervix,58 cases of cervical intrepithelial neoplasia (CIN), 39 cases of primary cervical squamous carcinoma .Results: (1) The expression of KAI1 tended to decrease with tumor progress (P<0.05). Expression of HPV tended to increase with tumor progress (P<0.05) . (2) The expression of KAI1 protein was decreasing with descending of cell histological grade in cervical carcinoma (P<0.05) .But the expression of KAI1 in different patients'age ,clinical stage and lymphonode metastasis had no statistical difference (P>0.05) . Expression of HPV statistically had nothing to do with patiants'age ,clinical stage, histological grade and lymphonode metastasis . (P>0.05). (3) In cervical carcinoma . the expression of metastasis suppressor gene KAI1 was decreased or absent , while the expression of human papillomavirus type 16/18 E6 and type 16E7 were getting significantly stronger progressively (P<0.05).There was no significant correlation between the expression of KAI1 and HPV (P>0.05).(4) Expression of HPVE6 tended to increase with tumor progress (P<0.05) .There had significant difference between control group and CIN, CIN and cervical carcinoma (P<0.05) . Expression of HPVE7 tended to increase with tumor progress (P<0.05) .There had significant difference between control group and CIN, CIN and cervical carcinoma (P<0.05) . The positive correlation between the expression of HPVE6 and HPVE7 was observed in cervical carcinoma .Conclusion: KAI1 might play an important role in the occurrence of cervical carcinoma . The decrease of KAI1 expression might be used as a prognostic marker which indicates the occurrence of cervical carcinoma . Human papillomavirus play an important role in the occurrence of cervical carcinoma .
