Bile duct carcinoma is one of the most malignant cancers, the 5-year survival is only 10%. The radical excision is still the main methods to treat Cholangiocarcinoma, but the therapeutic efficacy still don′t reach to an ideal condition. And moreover, there are about 30% patient have no chance to operate when they go to see a doctor. Therefore, one of the most hot topic in the study of Cholangiocarcinoma is to go deep into the mechanism and find new methods to improve the therapeutic efficacy.Objectives The aim of the study was to investigate the expression of the NF-κB and to explore the possible mechanisms by which it effect carcinoma of bile duct.Methods Patients: embedded specimens from 56 patients who underwent operation for bile duct carcinoma in the first affiliated hospital of Shan Xi medical University from 2000 to 2005 were selected for this experiment. There were 31 male and 25 female. The mean age was 55 years (32-74). All specimens were adenocarcinoma, and well-differentiated adenocarcinoma 18 cases, moderated-differentiated 26 cases, poor-differentiated 12 cases; with lymph nodes metastasis 25 cases; with nerve invasion 18 cases. The control cases were from benign lesion tissue of extrahepatic bile duct.Results The positive rate of NF-κB was 82.14%(46/56) in Cholangiocarcinoma and only 18.92%(7/37) in benign lesion tissue of extrahepatic bile duct. There is significant difference between two results(P<0.01). The positive rate of VEGF and cyclinD1 was 78.57%(44/56)and 73.21%(41/56)in Cholangiocarcinoma, while 40.54%(15/37),21.62%(8/37)in extrahepatic bile duct tissue. The positive rates of VEGF in Cholangiocarcinoma was higher than normal bile duct tissue(P<0.01), as cyclinD1 was. The expression of NF-κB was no associate with age, sex and mass location, while associated with histological type(P<0.01). Statistically significant difference in the expression of NF-κB could be observed between patients with positive and negative lymph node metastasis ,also between patients with positive and negative nerve invasion(P<0.01). The correlation coefficient was 0.590 and 0.683 between NF-κB with cyclinD1 and VEGF that show great significance(P<0.01).Conclusion1,The expression of NF-κB is higher in bile duct carcinoma and has closely relation with the occurrence, development , invasion and metastasis of Cholangiocarcinoma.2,NF-κB plays an important role in the pathogenesis of Cholangiocarcinoma by...
