For the purpose of the crackdown of the counterfeit drugs, a "Fast Drug Identification System", which includes a NIR prescreening identification system, a fast chemical identification system and HPLC-MS, is being developed by NICPBP to meet identification needs at different levels. This thesis emphasized on the study of the common rules of constructing the NIR ident model and the fragmentation rules in ESI-MS for β -lactam antibiotics. For the NIR prescreening identification system, the common method for constructing qualitative model, which included the optimization of the wavelength range and data preprocessing, selection of the factor combinations and the principle of determining the thresholds, was summarized by constructing a universal NIR qualitative model for identification of 26 kinds of cephalosporin for injections. Then two qualitative models for penicillin and aminoglycosides antibiotic for injections were constructed respectively to supply and validate the method. The accuracy, specificity and transformation of the models were ideal, and these models are now be used in drug identification vehicles. For the ESI-MS library, the fragmentation rules of β -lactam antibiotics in optimized condition were studied in an ESI ion source after optimizing the parameters of the ESI-MS, and confirmed by ESI-MS/MS. Then ESI-MS libraries for cephalosporin and penicillin antibiotics were established at the foundation of these experiments. It is an important complement to the retention value from HPLC and can be used for fast identification of drugs.
