To investigate the mechanism action of how the active ingredients from Ailanthus altissima prevented ulcerative colitis(UC). In this experiment, at first, a kind of water extraction and immersion ointment powder in a water system, and four kinds of ethanol extraction such as 95%ethanol extraction, 65%ethanol extraction, 45%ethanol extraction, 25%ethanol extraction were made in different ways, using water and ethanol as a solvent. Then, mice were used as the experimental animal and they were divided into eight groups by random, that is, normal group, model group, water extraction group, water+ethanol group, 95%ethanol extraction group, 65%ethanol extraction group, 45%ethanol extraction group, 25%ethanol extraction group.The ulcerative colitis(UC) model in mouse was made be 2 , 4-Dinitrochloro- benzene(DNCB), after being estimated successfully by the criterion, all kinds of extraction were injected into the stomach of the model mice by 150mg·kg -1. Seven days later, the action in pathological morphology of all kinds of extraction to UC mice was estimated by observing physical sign, structure variety characteristics of pathologic anatomy and pathological histology, meanwhile, immunological and biochemistry techniques were used to determine levels of MPO and PGE2 in colon tissue, the levels of NO,NOS,SOD,MDA lytic enzyme and levels of lytic enzyme in spleen.The results showed that: the water+ethanol,45%ethanol group, 65%ethanol group and 25% ethanol group of Ailanthus altissima could increase markably the weight of UC mice and reduce the number of mice with diarrhea and hemaferia and it's mortality. The results also show that the things above notably amend the mice's colon histological damnification and transformation in pathology, meanwhile, it was discovered that they can stimulate the immunological action of of spleen and thymus, and strengthen the function of immunology. However, water extraction and 95%ethanol extraction didn't have such function. Besides, different extraction of Ailanthus altissima could differently magnify the Spleen Index and Thymus Index of UC model mice. They could effectively restrain the activity of MPO in colon histology of UC mice and reduce the level of PGE2, meanwhile, they could also decrease NO free radicals in UC mice, strengthen the activity of SOD, clear MDA which was the production of skin twice oxidation reactions. They could variably enhance the level of ADA in serum of UC mice, lytic enzyme and ADA in spleen simultaneously. However, their degree varies much, in them, the effect of water+ethanol was the visiblest, water extraction was worse than other groups.Therefore, the extraction from Ailanthus altissima could differently protect and repair colon histology of UC model. Furthermore, it was probably one of the important mechanism of this action that their extraction could prevent some inflammation mediator in colon histology of UC mice, clear NO free radicals and oxygen free radicals, decrease degree of resisting skin bioxidation reactions.
