Epilepsy is a disorder characterized by recurrent episodes of paroxysmal brain dysfunction due to a sudden, disorderly, and excessive neuronal discharge.The particular cellular and molecular mechanism results in this syndrome is multiple and complicated,and there is still much manifest beyond our understanding. Astrocyte is one of the hot spots of research because of it's synthetic and secretory fuction. To investigate the effect of astrocytes on the genesis and development of epilepsy,as well as to probe the mechanism during this process, we performed the following experiments.In the first stage,we adopted pentylenetetrazol (PTZ) kindling method and successfully established a chromic epilepsy kindling model. In continuous operations ,we specifically observed the alteration of rat hippocampus with epilepsy and investigated the effects of estrogen and it's receptor antagonist on them.1. Establishment of a rat kindling model of epilepsyRat kindling model of epilepsy was established by using low dose(35mg/kg)PTZ intraperitoneal injection. We suggested that the method was an available and relative safe way avoiding the activation of astrocytes caused by surgical trauma. Subsequently,we used Racine's method to evaluate the rats and finally choseⅣtoⅤlevel of seizure ones to continue the research.2. Effects of Estrogen and its receptor antagonist on Astrocyte and Synapsy in Hippocampus of Kindling-model RatsIn this study, Male rats for experiment were randomly divided into normal control group,none -interfered group , estrogen–interfered group and estrogen +Tamoxifen(mixed estrogen antagonist) group.The model groups were treated by PTZ-kindling method and the behavior of rats in each group was observed.The expressions of GFAP and synaptophysin were determined by immunohistochernical technique in the hippocampus of the rats. Our data showed that Epileptic seizure occoured earlier and more severely in most rats of estrogen-interfered group than non-estrogen-interfered group . Rats in both model groups...
