Objective We analyze seizures and hippocampal sclerosis through establishing amygdala kindling model and using electrophysiological, immunohistochemical techniques. From both sides we study the c-Jun N-terminal kinase (JNK) signaling pathway and the effect of the JNK specific inhibitor SP600125 during the formation of epilepsy in the body. To explore the formation mechanism of epilepsy, ultimately, clinical medicine and surgery for the treatment of refractory epilepsy has opened new avenues.Methods We implant five core sites of microelectrodes into the right amygdale and place catheter in the left lateral ventricle of Wistar rats by stereotactic surgery. A blank group, a control group, and an injection drug group, were set up. On the eighth day after surgery, ventricle injection and electric stimulation were delivered to reach the kindling threshold. The performance in rat behavior and the EEG of frontal cortex were recorded during the whole kindling process. The rats were decapitated after the success (RacineⅤgrade continuous attack 10 times) in the kindling model of epilepsy. The rat's brains were made into paraffin section. H-E staining, Nissl staining, GFAP staining. They were analyzed to detect the cases of seizures, the histological changes of hippocampus and amygdala region.Results The mean time about the injection control group rats fully kindled under the 0.5 milliampere electric current is 21.79±2.29days. The time of injection drug group is 28.57±2.31days. The time of the injection control group rats was significantly shorter than the injection drug group. The changes found in H-E staining, Nissl and GFAP staining tissue slices: The neurons in stratum pyramidale of the hippocampus were showed depletion and the glia cells were displayed hyperplasia. The change of the injection group was significantly better than the control group.Conclusions1. The pathological changes of the hippocampus in the kindled rats are similar as it in the human mesal temporal lobe epilepsy: The neurons in stratum pyramidale of the hippocampus appear deletion and the glia cells were displayed reactive hyperplasia2. SP600125 has some cytoprotection for neuron in the epileptic seizure. The c-Jun N-Terminal Kinase signaling pathway take parts in hippocampus sclerosis in mesal temporal lobe epilepsy.
