As we all know,Multidrug Resistance(MDR) of leukemic cells is one of the major factors leading to chemotherapeutic treatment failure and relapse of Acute myelocytic leukemia(AML). Up to now, there is no effective and satisfactory measure to reverse MDR, as a result, early diagnosis is urgently needed, and providing early diagnosis is also another effective way to reduce and prevent the arise of MDR. The overexpression of multidrug resistance gene is the main mechanism of MDR, so the study of how to detect expression of MDR gene mRNA quickly and accurately has been a focus of attention in the field of leukemia.Reverse Polymerase chain reaction(RT-PCR) technology is one of the molecular diagnostic methods that has been developed in detecting the expression of MDR gene mRNAs. It has proven to be a quick method for identifying the presence of mRNA. But the genetic complexity of this non-target nucleic acid may also generate the amplification of a false positive product when a primer anneals to a mismatched sequence,especially as the number of cycles increases. And the specificity of the test is often inadequate,which will limit its applicable field. The technologies of immunohistochemistry and flow cytometry also have similar limitation in detecting the MDR.Using gene chips can resolve these problems. Gene chips have become one of the most powerful tools in the area of gene diagnostics and in the genetic analysis of biological samples. Gene chips also named as DNA microarray, which will provide researchers with the opportunity not only to detect the MDR gene mRNA expression in AML in a more sensitive, specific, speedy, efficient and automatic manner, but also to detect and genotype MDR gene simultaneously. We proposed to set up a rapid experimental procedure based upon the gene chips technology, in a hope to detect the MDR gene mRNA expression quickly and accurately.
