Background: Invasion and metastasis are two major properties of cancer, and metastasis is responsible for the majority of failures in treatment and most cancer deaths. The processes of metastasis include growth of the primary sites, angiogenesis, degradation of the ECM, intravasation, circulation, adhesion to endothelium of microvascular, intravascular proliferation, extravasation and growth in the second sites. The adhesion between cancer cells and endothelium of target organ is one of the key step in metastasis. But most previous experiments in vitro by cell culture of endothelium and cancer cells are deficient of authenticity and objectivity because of no influence of microenvironment and haemodynamics in vivo. This process and molecular mechanism of adhesion have been studied more directly and researched deeper than before due to the invention of fluorescent microscopy and the method of IVVM( Intravital videomicroscopy), which provide novel method to study metastasis, playing an important role in clinical research.Objectives: To establish the model of tumor cells attached to endothelium of pulmonary microvessels in early metastasis, by which to study the location and modalities of endothelium-attachment tumor cells in early metastasis to the lung.
