Objective:To observe endothelial damage lipid abnormality in rabbits and hawthorn leaf extract intervention role, explore the hawthorn leaf extract on the possible mechanism of vascular endothelial injury.Methods:The experiment system we selected is rabbit,50,male, weights 1.9-2.3kg.10 of them were selected as the normal group, which were fed with common fodder 150g/day per animal. Others were duplicated by feeding high lipid fodder (1%cholestero,5%custard powder,5% lard,89% common fodder). Four weeks later, all rabbits were forbidden to feed for 12 hours while given enough water.TC,TG,LDL,HDL were tested. The result of TC was higher fiver times than that in the normal group. And it is judged as that Dyslipidemia of rabbit model was built. Then based on the health condition of the animals,30 rabbits were divided into three groups, including model group,positive control group,extract of hawthorn leaf group. Then, the rabbits in positive control group were treated with Xuezhikang while which in the extract of hawthorn leaf group were treated with extract of hawthorn leaf. The diet was the same as before.Four weeks later,blood lipids level as well as the content of MDA and SOD were detected by RIA kit.VCAM-1 and VEGF were detected by ELISA. The expression of NOS was detected by IHCA.Results:The TC,TG,LDL-C,MDA in positive control group were decreased while the SOD and NOS were increased, compared with the model group(P<0.01 or P<0.05). The result of ELISA showed that Xuezhikang could decrease the VCAM-1's and the VEGF's content((P<0.01).The level of HDL-C was increased in positive control group. But no statistical significance compared to the model group.The results of Shanzhaye group were similar to the positive control group.Conclusion:Dyslipidemia can bring antioxidant capacity in rabbits reduced, causing lipid peroxidation and injury of dyslipidemia rabbit's endothelial cell. And it caused the degradation of endothelium-dependent relaxing function, which was at risk of AS. After 4 weeks treatment of Xuezhikang or extract of hawthorn leaf, lipid metabolism and anti-lipid peroxidation were improved a lot, which may bring about therapeutic effects on endothelial cell.
