Chitosan is recommended as suitable functional materials, because these natural polymers have excellent properties such as biocompatibility, biodegradability, non-toxicity, adsorption properties, etc. Chitosan is the N-deacetylated derivative of chitin, although this N-deacetylation is almost never complete. A sharp nomenclature with respect to the degree of N-deacetylation has not been defined between chitin and chitosan. Recently, much attention has been paid to chitosan as a potential polysaccharide resource as the new medicine complement. Floating drug delivery system (FDDS) is one of the approaches to increase the retention time of an oral dosage form in the stomach. FDDS have a bulk density lower than gastric fluids and thus remain buoyancy in the stomach without affecting the gastric emptying rate for a prolonged period of time.Sodium ferulate is the sodium salt of ferulic acid that is an effective component derives from the Chinese traditional medicinal materials, such as Radix Angelicae Sinensis and Rhizoma Chuanxiong.Water uptake and swelling of the floating tablet is the keys to the buoyancy generation and duration of floating drug delivery system. The study on the relationship of water uptake and swelling demonstrated that in all cases swelling expansion can generate a buoyancy force substantial enough to counteract water-uptake increases.This study for gastric retention was made by using sodium ferulate as a model drug, which is effervescent FDDS on the way of direct press. Chitosan was the main matrices, HPMC was the second matrices, and stearyl alcohol was bleaching auxiliaries while NaHCO3 is the effervescent formulation. On basis of pre-experiments and pharmaceutical pre-formulation studies, the amount and types of excipients, prepared methods and drug release in vitro of the prescription were determined by buoyant and drug releasing conditions as screening index with orthogonal design test. Drug releasing process of testing tablets in media was accorded to Peppas equation, and the releasing mechanism was non-Fick's diffusion. The study of forepart stability indicated that the preparation should be stored in the dry a light-prevented environment especially in the moist.The study of hemorheology indicated that sodium ferulate floating tablets and the common tablets had a certain effect to blood viscosity, plasma viscosity, Hct and ESR,...
