The Study Of AT1R Blocker On The Growth And Angiogenesis Of Human Ovarian Cancer Transplanted Subcutaneously In Nude Mice

Objective To study the growth of Losartan on HO8910 in vitro and the effect of losartan in the growth and angiogenesis of neoplasm in nude mice implanted subcutaneously with human ovarian cancer cell (HO8910) in nude mice.Methods 1.The effects of different concentrations of losartan applied in HO8910 in vitro were observed by MTT assay 2. Human ovarian cancer transplanted subcutaneously in nude mice model was established,then divided into 4 groups: contral group, losartan groups(10 and 100mg/kg in body weight) ,and DDP group.Beginning on day 2, intragastric administration of losartan(10 , 100mg/kg in body weight) or NS were done daily and DDP was injected i.p.once a week. different treatments were served after trasplantation and all mice were sacrificed after 40 days .Tumor volume and weight ,VEGF protein, VEGF mRNA, MVD were measured .The level of VEGF mRNA in tumor tissue was determined by relative quantative reverse transcription polymerase chain reaction.VEGF protein and MVD was calculated by immunohistochemistry.Results 1. Losartan suppressed the proliferation of HO8910 in a dose- dependent manner.2.The tumor volume in losartan groups and DDP group was smaller than it in contral groups(p<0.05).3.Expression of VEGF protein level in losartan groups measured by immunohistochemistry was lower singnificantly than it in contral groups( p<0.05).MVD was obviously reduced in losartan groups(100mg/kg group and 100mg/kg group were 15.03 ± 3.12;19.71 ± 3.08 respectively) than in contral group(26.6 ± 4.34 , p<0.05).4.Expression of VEGF mRNA level in losartan 100mg/kg group (3.67 ± 1.27) was lower remarkably than in contral group (17.03 ± 4.84).Conclusions 1.
