Objective To study the growth of thalidomide with the combination of cisplatin,5-fluorouracil and etoposide on SKOV3 in vitro and the effect of thalidomide used alone and in combination cytoxan (CTX) in the growth and angiogenesis of human ovarian cancer transplanted subcutaneously in nude mice.Methods 1.The effects of different drugs of concentrations were observed by MTT assay .The VEGF content of SKOV3 in culture medium was tested by ELSIA,coefficient of drug interaction was used to analysis the synergistic effect of the drugs. 2. Human ovarian cancer transplanted subcutaneously in nude mice model was established,then divided into 3 groups:contral groups,Thd groups ,Thd+CTX goups,different treatments were served from day 8 after trasplantation and all mice were sacrificed after 28 days ,tumor volume and weight ,VEGF mRNA,VEGF protein,MVD were detected.The level of VEGF mRNA in tumor tissue was determined by relative quantative reverse transcription polymerase chain reaction.VEGFprotein level in serum was determined by ELSIA. MVD was calculated by immunohistochemistry.Results 1.There were effects in inhibition rate and proliferation rate after the different concentration Thd..The VEGF content of SKOV3 in culture medium with Thd was lower than it in control (p<0.01). Thd combination group could get the effect of synergistically inhibit(p<0.01) .2.The tumor volume in Thd groups and Thd+CTX goups was smaller than it in contral groups(p<0.05).Expression of VEGF mRNA level in Thd groups(55.04± 9.16) and Thd+CTX goups (26.43 + 7.4) was lower singnificantly than it in contral groups(78.67 ± 7.14, p<0.01).Serum VEGF level in Thd groups(29.44±10.03 pg/ml) and Thd+CTX goups (11.94 ±5.87 pg/ml) was lower singnificantly than it in contral groups (p<0.01).MVD was obviously reduced (p<0.01).Conclusions l.Thd can synergistically inhibit the proliferation of SKOV3 with combination of cDDP,VP16 or 5-FU,which can give reference in chemotherapy.2.Thalidomide can inhibit the growth and angiogenesis of human ovarian cancer transplanted subcutaneously in nude mice.Treatment with thalidomide is a potentially useful methed of antitumor therapy in ovarian cancer.
