Objective: To detecte the chromosomal aberration in the tissue cells of lung cancer and to evaluate the relationship between the chromosomal aberration and clinical features of lung cancer.Meterials and Methods: Flow cytometry(FCM) was used to analyze the DNA ploid disposition in the fresh cancer tissue cells of 30 patients with lung cancer,out of whom,17 were performed in comparative genomic hybridization to detect the global genomic aberration in cancer tissue cells.Rustlts: 1. DNA heteroploid was found in 19 cases(63.3%).DNA heteroploid disposition has no relation with pathological type,the status of lymph node metastasis and clinical stage of patient.2. All of 17 cases detected by CGH showed chromosomal aberrations.Each case involved with a few chromosomes, from 4 to 24.The average numbers of chromosomal gains and losses in each case in NSCLC were 7.0 and 4.8 respectively; corresponding to 8.4 and 9.6 in SCLC. The frequencies of gains and losses on chromosome has no significant differences between NSCLC and SCLC.3. The regions and frequencies of chromosomal aberration in NSCLC were different from that in SCLC. In NSCLC, the regions and frequencies of chromosomal gains in turn were 1p(75%,9/12), 3q24-28(58.3%,7/12), 11q13(58.3%,7/12), 19q12-21 (58.3%,7/12), 5p12-15(33.3%,4/12), 8q(33.3%,4/12), 14q23(33.3%,4/12), 20p12-ter (33.3%,4/12), 21q21(33.3%,4/12) and 4p13(25%,3/12). In SCLC, the regions and frequencies of chromosomal gains in turn were 1p(100%,5/5), 20p12-ter (80.0%,4/5), 19q12-21(60.0%,3/5), 5p12-15(60.0%,3/5), 21q21(60.0%,3/5), 8q(40%,2/5) and 4p13 (40%,2/5).Among them, the frequencies of gains on chromosomal arms 3q24-28 and 11q13(58.3% and 58.3%) in NSCLC were significantly higher than that in SCLC(0% and 0%)(p<0.05 and <0.05,respectively).
