Objective: To characterize the profiles of chromosome imbalance (loss and gain) of Xinjiang Kazakh's primary esophageal squamous cell carcinoma(ESCC).And to analyse the relationship between the genetic aberrations and the clinical characteristics as well as infection of HPV.Methods:(1)Comparative genomic hybridization (CGH) was used to examine 54 cases of Xinjiang Kazak's primary ESCC based on the result of HPV infection which has been detected by our team-workers ,the result of CGH were analysed by the specil software of METASYSTEMS desined by Germany.And All of the data were processed with the SPSS 13.0 software package (SPSS Inc., Chicago, IL).Results: Fifty-four cases of Xinjiang Kazak's patients with esophageal squamous cell carcinoma(ESCC) showed evidence of increased or decreased DNA sequence copy numbers involving one or more regions of the genome. In total, 683 gains and 488 losses were found in 54 tumor samples; thus,the average gains and losses per patient were12.64 and 9.04, respectively. (1) Frequent gain abnormalities were found on chromosome arms 8q(35/54,65%),1q(32/54,60%),3q(30/54,56%),18q(29/54,54%) ,5p(28/54,52%),11q(26/54,48%),13q(24/54,44%), Frequent deletions were found on chromosome arms 22q(28/54,52%), 8p(24/54,44%),13q(21/54,39%),17p(21/54,39%),among them the common region of gains and loss are 8q22-24(32/54, 60%),1q13(29/54,54%),3q21-22(27/54,50%),18q21-24(22/54,42%),5p13.1-ter (16/54,30%)and22q11.2-13.3(26/54,48%),8p22-ter(19/54,35%),13q13-21(18/54,33%),17p13(16/54,30%), respectively;(2) It was found that deletions of 3p,17p,22q and gains of 5p,11q were significantly correlated with the location of tumors. Losses of 1p,3p,22q and gains of 3q,8q,18q also were found more frequently in patients with larger diameter disease. Losses of 19q and Gain of l3q,18p were significantly correlated with pathologic grade (p<0.05),among them , gains of l3q were positive correlated with pathologic grade ,loss of 19q was negatively correlated with pathologic grade.The gain of 7p and deletion of 19q was seen more frequently in patients with Gradeâ…¢-â…£tumors.Gains of 1q,7p,8q and losses of 3p,11q,17p were significantly correlated LN metastasis.The more frequent aberrations were seen in patients with LN metastasis.(3) gains of 1q,6q and loss of 11q were significantly correlated with the type of HPV infection.The gain of 1q was possible distinct changes of HPV16, gains of 6q and loss of 11q were distinct changes of HPV18,chromosomal aberrations were significantly correlated with the type of HPV infection.Conclusions:These observations suggest that (1) the gains and/or losses of genomic DNA from Xinjiang Kazak's squamous esophageal carcinomas are the common molecular cytogenetic aberrations; Gains in part or in whole of chromosome8q,1q,3q, 18p, 5p, 11q, 13q,18p,20q and losses of 22q,8p,13q,17p,5q,10q,19q were detected frequently in Xinjiang Kazak's SCC.these genetic alterations may correlated with Kazakh's ESCC-related carcinogenesis (2) Gain of 7p may correlated with poor prognosis, and loss of 19q may correlated with the progression of disease.(3) Infection of HPV mybe involved in the disease progression of Xinjiang Kazak's EC-SCC.in light of the association of chromosomal imbalance with HPV.among them, the gain of 1q mybe be considered as the characteristic of HPV16 infection , the gain of 6q and delection of 11q mybe be considered as the characteristic of HPV18 infection, further reseach should be pay more attention.
