To observe the influence of dexamethasone (DEX) and triamcinolone acetonide (TA) of different concentrations on cells growth and vascular endothelial growth factor (VEGF) expression of cultured human retinal pigment epithelial cells (hRPE).The methods are as follows: human retinal pigment epithelial cells were culturd in vitro. In the first part of the experiment, different concentration (1,10,50,100,200,400μg/ml) of DEX were added to the cells, with the contrast of vacuity. The cells were takend into two parts and were culturd in normal (95% oxygeon) and hypoxia(2%-5%oxygeon) condition for 24 hours. The influence of dexamethasone on cells growth and VEGF expression was detected by 3- (4,5-dimethylthiazole-2yl) -2,5diphenyl tetrazolium bromid (MTT) and Reverse transcription (RT-PCR ) assay. In the second part of the experiment,we added different concentration (10,100,400,800,1600μg/ml) of TA to the cell, with the contrast of vacuity. The rest condition was the same of the first part. The data were collected and analysed with independent sample T test and One Way ANOVA.In the first part of DEX experiment ,the MTT shows that A values of cells at hypoxia condition were higher than those in normal condition at the same drug concentration (P<0.01 ) .There were no obevious significant between...
