Object i ve: (1) To research and develop a photochemical methed that can inactivate virus using riboflavin plus UV light. (2) To investigate the inactivation effects of UV light plus riboflavin for virus and bacteria in plasma.. (3 ) To study the influences on clotting facter activity of disinfected plasma by riboflavin plus UVlight. Metheds: (1)Using Vesiculer Stomatitis Virus (VSV) as a model virus. Adding riboflavin (0,50,100,200,300μmol/L) to VSV(7.25 lg TCID50)in PVC blood bag and then the mixture was exposed to ultraviolet light ( λ : 250-350nm) for 0, 5, 10, 20, 30, 60 minutes, Mixture of riboflavin and virus suspensions treated with different riboflavin concentration and illuminating time was collected and inoculated in single layer cells. And the virus titer was measured. The inactivation effects of UV light plus riboflavin for VSV was stuied.(2)In this study,Vesiculer Stomatitis Virus (VSV ) , Escherichia.coli and Staphylococcus.aureus were used as model virus and model bactiria. Adding riboflavin (0,50,100,200,300 μ mol/L) to VSV(5. 93 lg PFU/mL) in PVC blood bag which exposed to ultraviolet light ( λ: 250-350nm) for 0, 5, 10, 20, 30, 60 minutes, And then the virus titer was measured by Plaque Forming Units Assay(PFUA). Mixture of riboflavin and bacterium suspensions treated with different riboflavin concentration and illuminating time was collected. The inactivation effects of UV light plus riboflavin for virus and bacterium was stuied.(3)The best inactivation of this methed was screened by Orthogonal assay, after the best inactivation has been chosen, the activation of clotting facter Ⅴ,Ⅷ,Ⅹ and...
