ObjectiveTo investigate the herbs containing acid on mechanism of synthesis of melanocyte at the molecular and biochemistical level, and testify the heibs containing acid could inhibit Synthesis of Melanocyte and provide experimental evidence for pursueing effective natural drugs to prevent and treat the disease of pigmentation. Methods1.Looks up the literature to pick out 28 kinds of the herbs containing acid, Separately extract acid through method of alkaline water, tyrosinase inhabitory activity in vitro was determined by L-DOPA oxidiziation and choose the highest rate of inhabitory activity from the results to make the further study on pigmentation of skin with UVB.2.Pigmentation models were induced by UVB. Guinea pigs(female ,pure white) were involved in and randomly divided into normal control group, model control group, menstruum control group , and dark plum high, middle and low groups, as well as positive drug group. Except for the normal control group, the other six groups of hair were cut and were daily exposed for 30 min/day up to 4 consecutive weeks to establish the pigmentation models and use drugs for four weeks. we had observed the crude extracts from dark plum to influence the skin of guinea pig on pathomorphism and the expression of NOS Results1.28 kinds of crude extracts in high density (25mg/Ml) showed potent inhibitory action on tyrosinase activity. The highest inhabitory rate is 96.50%and the extracts of organic acid is from dark plum2.Two kinds of crude extracts (dark plum 96.50%, shizandra berry85.53 %) showed potent inhibitory action on tyrosinase activity compared with positive control arbutin (2mmol/ml, P< 0.01;) Eight kinds of crude extracts (fruit of belvedere 81.66%, loquat 81.65%, rose-hip81.12%, cherokee selfheal 77.51%earthworm77.09%, stigma of corn 66.74%, chuanxiong66.31%, myrabolam 62.03%,) showed potent inhibitory action on tyrosinase activity compared with positive control arbutin (2mmol/ml, P< 0.05).3.Twenty kinds of crude extracts in high, middle and low density showed potent inhibitory action on tyrosinase activity and the effectiviniss of inhibitory was related with their concentration. Eight kinds of crude extracts showed potent inhibitory action on tyrosinase activity in high density and activation in middle and low density.
