Objective: To detect the ratio of CD34 positive cells in bone marrow mononuclear cells (BMMNC) and the expressive rate of granulocyte-macrophage colony-stimulating factor receptors (GM-CSFR, GMR) on CD34 positive cells of the patients with aplastic anemia (AA) and myelodysplastic syndrome (MDS), So to discriminate the difference of the two diseases in the hemopoietic stem/ progenitor cells and molecular level, and to reveal the pathogenesies of AA and MDS , that help us not only diagnose and distinguish AA and MDS but also evaluate prognosis in clinic.Method: 14 AA patients, 23 MDS patients and 8 normal subjects were respectively taken 2ml bone marrow samples by conventional bone marrow puncture. After separating bone marrow mononuclear cells with gradient centrifugation, to detect the ratio of CD34 positive cells in bone marrow mononuclear cells and the expressive rate of GM-CSFR on CD34 positive cells by flow cytometry (FCM) and monoclonal antibody technique.
