Objective The aim of the present study was to investigate the effect of paeoniflorin on acute myocardial ischemia in rats and dogs.Methods Acute myocardial ischemia was prepared by pituitrin in rats and ECG indexes were observed. Myocardial ischemia was induced by coronary artery ligation in anaesthetized dogs, and then ECG was recorded, AST, CK, LDH, SOD and MDA in serum were determined respectively, the myocardial infarction area was measured by nitroblue tetrazolium (NBT) dyeing. Cardiac hemodynamic parameters were examined in anesthetic chest-opened dogs with various doses of paeoniflorin.Results T wave in rats changed significantly after pituitrin intravenously, which was reversed by paeoniflorin. Paeoniflorin decreased the infarct size dyed by NBT, attenuated the degree and range of myocardial ischemia in experimental myocardial infarction canine model, and the activity of creatine phosphokinase (CPK), the MDA level were also decreased by paeoniflorin, but the activity of SOD was increased. Paeoniflorin decreased systolic blood pressure(SBP) , diastolic blood pressure (DBP), left ventricular systolic pressure(LVSP), left ventricular work index (LVWI) and ± differential pressure/differential time maxium (±dp/dtmax).The heart rate and the myocardial oxygen consumption was reduced. The total peripheral vascular resistance (TPR) and coronary arterial resistance (CAR) were reduced, then the afterload of heart was decreased, resulting in increase of heart perfusion flow.
