Objective To study the effect and machanism of photocatalyst TiO2 on bovine lens epithelial cells irradiated by ultraviolet-A(UVA) and evaluate the possibility of preventing posterior capsule opacification(PCO) .Methods Culturing the bovine lens epithelial cells with different concentrations of TiO2, we measure its influence on cell growth rate and cells cycle by the ways of MTT Colorimetric Assay and the flow cytometry ,and study its photoexciting effect by the ways of MTT Colorimetric Assay, Trypan Blue Exclusion, AO/EB (Acridine Orange/Ethidium Bromide) dyeness, DNA agarose gel electrophoresis and electron microscope.Results After the bovine lens epithelial cells were cultured with different TiO2 concentrations, TiO2 particles were found to enter into the cytoplasm via phagocytosis. The growth rate of the bovine lens epithelial cells was decline with the increase of TiO2 concentrations. The most significant inhibitory effect of growth was in the presence of 200ug/ml TiO2 for 48 hours, while the cells cycle show the decline of proliferation index. There was significant difference between TiO2 group and the control group(P=0.001). The photoexciting effect of TiO2 was correlated with the concentration of TiO2 and irradiation time.
