Background Tumor is the common ailment which affects serially medical fitness of human, so it is the hot topic for medical research. It is widely acknowledged that tumor progression and metastasis is a highly complex process involving a variety of factors. And the disease is concerned with alteration of genetic constitution and function and/or difference of genetic expression and regulation. Lots of genes have been certified to correlate with tumor so far, however, it can't elucidate the mechanisms of tumor progression and metastasis completely. Adenoid cystic carcinoma (ACC) is characterized by its high rate of metastasis and perineural invasion. The research about gene expression in ACC metastasis will contribute to reveal the mechanisms. Recently, restriction fragment differential display PCR (RFDD-PCR) which based on mRNA differential display reverse transcription PCR (DD-PCR) has been developed as a new technology. Depending on reliability of restriction fragment length polymorphism (RFLP) and high efficiency of PCR, the technology can amplify selectively to cDNA fragments and diminish the workload. It is a good approach for the identification of differentially expressed genes in a couple of cell colony and several samples. The technology had good reproducibility and coverage. Furthermore, it cut down the false positive rate by amplifying cDNA restriction fragments under stringent PCR conditions.
