| Objective: Investigation of the Kangnaoling granulation's protective effect on cerebral vascular injured rat's hippocampus neural cell of pasting cerebral pia mater with FeCl3 solution. Methods: We divide the rats into 5 groups in random, with Morris method, detect the change of some active oxygen radicals such as super-oxide demitasse (SOD), Nitric Oxide Synthase (NOS), malondialdehyde (MDA) etc. Also, we detect the change of cholinesterase (CHE), Monoamine Oxidize (MAO); using HE,PAS staining, immuno-histochemical technique, detect hippocampus CA1 cerebral pathology slices optical density value of NOS, CHE and lipofuscin of neural cell; detecting apoptosis rate and the expression of P53 , which are neuronal apoptosis related genes.Result Vascular Dementia (VD) model rat displays memory function decrease, blood vessel straitness or obstruction, related brain section neuron lost or distorted; cerebella lipofuscin, LPO increased; free radical injury especially. The results indicate that the KANGNAOLING granulation can promote the model rats' NO level, NOS activation and can improve the cerebral blood flow, decrease the further damage of the neurons. It can improve SOD activation, lower the amount of MDA, reduce the vigor of CHE and MAO, then suppress production of free radical and delay neuronal cell death. The KANGNAOLING granulation can suppress the express of p53, so it can reduce the apoptosis of neurons.Conclusion The Kangnaoling granulation can activate blood circulation to dissipate blood stasis, expand blood vessel, antagonize the neurons' damage and apoptosis of the cerebral ischemia-reperfusion rats' model. It also can improve the metabolism of free radicals, readjust the disordered neurotransmitter. It can play a positive role on recovery of the VD. |
