Objectives: 1 TO investigate the correlation between expression of VEGF and KDR,VEGF-C and VEGFR-3 in different histopathlogical grade of astrocytoma . 2 The current research is designed to investigate the effect of angiogenesis of VEGF in different histopathological grade of astrocytoma and whether ERK plays a role in this signal pathway.Methods: 1 The expression of VEGF and KDR ,VEGF-C and VEGFR-3 were detected by immunnohistochemically in 50 surgical specimen with brain astrocytoma. The microvessel density in astrocytoma intratumoral were examined by immunnohistochemically withâ…§factor . 2 Two cell sublines of human glioma SWO38Z cell lines SWO38Z1 and SWO38Z2, which have different differentiation, were used in this study. The expression of VEGF in SWO38Z1 and SWO38Z2 were detected by Western blot. 3 The effect of vascular endothelial cell,s proliferation with VEGF was measured with MTT assay. 4 Western blot is used to detect the expression level of vascular endothelial cell,s ERK1/2 phosphorylation in VEGF treated group and control group.
