objective: To research the loss of PTEN ,the relationships between protein expressions of PTEN,HIF-1α,VEGF, and correlations between the three and some clinicopathological characteristics in astrocytoma. Methods: Loss of PTEN was detected by RT-PCR; protein expression of PTEN,HIF-1α,VEGF were checked by immunohistochemical method. Results: (1) Four of 32 cases showed the loss of PTEN, which all belonged to the fourth grade. (2) 62.0%,57.7% and 64.8% were positive for PTEN,HIF-1α,VEGF expression, respectively, which were significant in low- and high- grade astrocytoma. HIF-1αshowed intensively around the border and the necrosis areas of tumors. (3)There were negative correlations between PTEN and HIF-1α,VEGF, and a positive one between HIF-1αand VEGF. (4) The protein expression of HIF-1αVEGF had no correlation with age, sex and tumor size. The protein expression of PTEN had no Only PTEN had no correlation with age, sex and tumor size, but had a negative correlation of age. Conclusions: (1) Loss of PTEN is a molecular event during the malignant process of astrocytoma. (2)PTEN has effects on HIF-1αand VEGF, HIF-1αis a regulative factor to VEGF. All three involve in the tumor...
