The genesis and development of the neoplasm include multi-genes and complicated processes of multi-level control. Among these, the variation of the normal gene is the initial factor, as well as its core. As genetic mutations accumulate, the malignant transformation of normal cell will occur. p53 gene, as a tumor suppressive gene, significantly correlates with the occurrence of the tumor when it mutated; and also the VEGF, which plays an important role in the promotion to the angiogenesis and tumor stroma, has become one of the main objects while researching in the development and invasion of the glioma.Objectives: To inqure into the roles of p53 and VEGF which play in the prossesses of the astrocytoma occurrence, proliferation and invade, and also examine the relationships and correlations of p53 and VEGF with the malignant degree of astrocytoma and the clinical significance.Methods: Immunohistochemical staining (S-P method) was executed to detect the expressions of p53 and VEGF proteins in 60 cases of astrocytoma and 10 normal brain tissue cases. The relationships between the expression of p53 and VEGF and clinically pathological features, as well as the correlations between them were analyzed by the statistics.Results:1. There are no positive expressions of the p53 protein in the specimens of normal brain tissue, the cases of positive expression of the p53 protein are 23 in the 60 specimens of astrocytomas, it shows a significant diversity between the two groups (P<0.05); With the grade of astrocytoma advancing, the percentage positive expression for p53 protein is higher,the expression of the possitive protein in the low grade (gradeâ… andâ…¡) is no meaning,so is the high grade (gradeâ…¢and IV) (P>0.05), nevertheless, the statistic between the low and high is noticeable (P<0.05); Correlation analysis according to Spearman reveals a significant positive correlation between the expression intensity of p53 protein and the grade of astrocytoma (P<0.05).2. The general percentage positive expression for VEGF protein in the human astrocytomas cases is 56.66% (34/60), whereas it is all that indicate negative in the specimens of the normal brain tissue, it shows a markable diversity between the two groups (P<0.05); The percentage is 15.38% in grade I astrocytomas, while the percentages are 29.41%, 87.50% and 92.85% in astrocytomas from grade II to IV, respectively, While we can observe that as the grade of astrocytoma advances, the percentage positive expression for VEGF protein was higher, the expression of the possitive protein in the low grade (gradeâ… andâ…¡) is no meaning,so is the high grade (gradeâ…¢and IV) (P>0.05), however, the statistic between the low and high isnoticeable (P<0.05), so, we can differentiate the low grade from the high grade; according to Spearman correlation analysis reveals a significant positive relationship between the expression intensity of VEGF protein and the grade of astrocytoma (P<0.05), There is a significant correlation between the expressed intensity of the protein and the pathological grade; the more malignant, the more expression intensity.3. There is a positive correlation between the expression of the VEGF protein and the mutation of p53 in human astrocytomas, according to the Spearman relative analysis (P<0.05).Conclusions:1. There are no expressions of the p53 and VEGF protein in the normal human brain tissues.2. The results demonstrate the correlations between the expression and its intensity of the p53 protein and histopathological grading in astrocytomas. It is a typical feature that the p53 protein has excess expression in the malignant and low-cell-differentiation astrocytomas.3. There are relations between the expression and its intensity of the VEGF protein and histopathological grading in astrocytomas. The expression of the VEGF-protein shows the conditions of the vascularization in the neoplasms, which relates to the occurrence, progression and invasion of the astroma.4. There is a positive correlation between the expression of theVEGF protein and the mutation of p53 in human astrocytomas, the two might co-operate and regulate each other in the processes of occurrence and growth of the astroma,and also we might combine the positive expressions of the two proteins to improve the concision of the pathological diagnoses and to analyze the endings and malignancies of the neoplasm.
