Background and objective:As the other solid tumors, the development and metastasis of lung cancer depends on angiogenesis. New vessels have been considered important targets to inhibit the angiogenesis of tumor and to block the resource of nutrition and the passageway, which have been a new strategy to fight against cancer in the past few years. To establish lung microvascular endothelial cell in three dimensional culture, to study the effect and mechanism of Avastin, a humanized anti-VEGF antibody, on the lung microvascular angiogenesis induced lung cancer in vitro. To investigate in vivo the effects of Thalidomide and Avastin on the nude mice. Material:DMEM(Low carbohydrates type), FBS, Insulin, Transferrin, EGF (epidermal growth factor), Dexamethasone, ECGS(endothelial cell growth supplement) ,PI , mRnase , Mouse mAb to human CD31,Mouse IgG, Rabbit mAb to mouse with TRITC, TNP-170, Methylrosanilinium Chloride, PFA(paraformaldehyde), filter membrane and MW10000 dialysis-membrane, Super-clean bench, carbon dioxide incubation box, flow cytometry. inverted microscope, LXJ- â…¡ conventional centrifuge, eppendorf5810R centrifuge, microamount-sample injector. Fluorescence microscope, electronic analytical balance, eye scissors, nippers.
